A Selectfluor‐based Polonovski Rearrangement Leading to Novel Entities for Synthetic and Medicinal Applications

open access: yesChemistry – A European Journal, EarlyView.
Starting from carboxylic acids and Selectfluor‐derivatives, functionalized diazabicyclooctanes can be obtained via a novel Polonovski‐type rearrangement proceeding under mild reaction conditions. Besides their use in organic synthesis the unique structure and reactivity of the rearrangement products enables applications in medicinal chemistry ...

Mild C─F Activation of Azido(per)Fluoroalkanes

open access: yesChemistry – A European Journal, EarlyView.
Mild and selective activation of C(sp3)–F bonds in azido(per)fluoroalkanes using thiolates was developed, leading to novel functionalized azides. The reaction is initiated by nucleophilic attack of the sulfur atom to the terminal nitrogen of the azido group, followed by fluoride ion elimination.

A Unified Approach for the Synthesis of Conformationally Locked and sp2‐sp3 Fused Hybrids

open access: yesChemistry – A European Journal, EarlyView.
This study presents a systematic platform to build rigid sp2–sp3 hybrids via benzyne cycloadditions. Arene‑fused bicyclic oxa‐, aza‐, and carbocycles are converted into medicinally relevant amino acids and applied to Enalapril analogues synthesis. The hybrids combine aromatic interactions with 3D rigidity, tuning lipophilicity, pKa, and solubility ...

Relay Approach: A Convergent Synthesis of Key Fragments en route to (+)‐Neosorangicin A

open access: yesChemistry – A European Journal, EarlyView.
A selective synthesis of three key fragments of the antibiotically active (+)‐neosorangicin A is reported, providing a foundation for its convergent total synthesis and proof of strategy via a relay approach. ABSTRACT Herein, we report the synthesis of the three key fragments of the macrolide antibiotic (+)‐neosorangicin A, a compound exhibiting potent

Stereochemical Control in the Matteson Reaction

open access: yesChemistry – A European Journal, EarlyView.
The Matteson homologation is a highly versatile method for the stereoselective insertion of carbenoids into boronic esters. Through iterative application of the process, complex chiral molecules can be synthesized. The stereoselectivity of the reaction is determined by a chiral diol incorporated in the boronic ester.
