Directed evolution of enzymes at the crossroads of tradition and innovation

open access: yesFEBS Open Bio, EarlyView.
An iterative cycle of data‐driven enzyme optimization comprising four stages: genetic diversification of a template enzyme, expression of protein variants, high‐throughput evaluation, and machine‐learning‐guided redesign of the next variant library.

Ephemeropteran and Trichopteran Assemblages Vary Across a Subtropical Rainforest Altitudinal Gradient: Useful Indicators for Climate Change

open access: yesEcology and Evolution
The subtropical rainforests of eastern Australia are expected to be greatly affected by climate change, with several studies predicting an upward shift in elevational distribution for many groups of fauna and flora.

Humic Acid Composition and Characteristics of Soil Organic Matter in Relation to the Elevation Gradient of Moso Bamboo Plantations.

open access: yesPLoS ONE, 2016
Studying the influence of climatic and/or site-specific factors on soil organic matter (SOM) along an elevation gradient is important for understanding the response of SOM to global warming.

Evaluating the involvement of autolysosomes in the nuclear translocation of fluorescent proteins

open access: yesFEBS Open Bio, EarlyView.
Endogenously expressed fluorescent proteins can be degraded by autophagy and transported to cell nuclei via the nuclear pore complex. But in some cell lines, for example, HeLa cells which are positive for immunoreactivity of a receptor ligand, such as UCN I, in cell nuclei, fusion of autolysosome with the nuclear envelope is involved in the nuclear ...

Yeast Gcn2 retains activity following humanization of its auto‐phosphorylation region

open access: yesFEBS Open Bio, EarlyView.
Using Saccharomyces cerevisiae as a model to study Gcn2 activation and regulation is limited by the lack of antibodies detecting phosphorylated Gcn2. To overcome this, we engineered Gcn2‐HsC, a yeast Gcn2 variant recognizable by commercial anti‐human phospho‐GCN2 antibodies.

Ionomycin suppresses cancer cell growth by disrupting mitochondrial transcription

open access: yesFEBS Open Bio, EarlyView.
In this study, we identify a new function for the selective Ca2+ ionophore, ionomycin, as an inhibitor of mitochondrial transcription. Both total and nascent RNA analyses revealed that ionomycin treatment reduces the transcription of mitochondrial genes.

Evolution‐guided yeast complementation reveals functional differences in human PSPH variants

open access: yesFEBS Open Bio, EarlyView.
Ancient genomes can help guide which human genetic variants are tested experimentally. This study applies that idea to PSPH, a gene involved in serine biosynthesis, and uses high‐throughput yeast complementation to compare variant function. The findings reveal measurable differences among selected alleles and illustrate the value of evolution‐guided ...

) varies significantly along an elevation gradient

open access: yes, 2012
International audienceLittle is known about the potential effect of climate warming on phyllosphere fungi, despite their important impact on the dynamics and diversity of plant communities. The structure of phyllosphere fungal assemblages along elevation

Four simplified gradient elasticity models for the simulation of dispersive wave propagation [PDF]

open access: yes, 2008
Gradient elasticity theories can be used to simulate dispersive wave propagation as it occurs in heterogeneous materials. Compared to the second-order partial differential equations of classical elasticity, in its most general format gradient elasticity ...
