Sensitive detection of endo-1,4-β-glucanases and endo-1,4-β-xylanases in gels
Analytical Biochemistry, 1985A simple, highly sensitive zymogram technique for detection of endo-1,4-beta-glucanases and endo-1,4-beta-xylanases in polyacrylamide gels after electrophoresis or isoelectric focusing was developed. The detection employs transparent agar replicas containing soluble covalently dyed polysaccharides, hydroxyethylcellulose dyed with Ostazin brilliant red ...

Structural and functional properties of low molecular weight endo-1,4-β -xylanases
Journal of Biotechnology, 1997There are currently four crystal structures of low molecular weight endo-1,4-beta-xylanases (E.C.3.2.1.8), i.e. family G/11 xylanases, available at the Brookhaven Data Bank: 2 xylanases from Trichoderma reesei (Törrönen et al., 1994; Törrönen and Rouvinen, 1995) and one from Bacillus circulans and another from Trichoderma harzianum (Campbell et al ...

Substrate-Binding Site of Endo-1,4-beta-Xylanase of the Yeast Cryptococcus albidus
European Journal of Biochemistry, 1981The substrate-binding site of endo-1,4-beta-xylanase of the yeast Cryptococcus albidus was investigated using, 1,4-beta-xylooligosaccharides (1-3H)-labelled at the reducing end. Evaluation of the affinities of ten imaginary subsites by the method of Suganuma et al. [1978, J. Biochem.

Abstract In this study, an optimization of xylanase expression by recombinant Pichia pastoris was carried out with a 2 3 factorial design. The influence of initial cell density, methanol and yeast nitrogen base concentration was evaluated, and the cell density was found to be the most important parameter.

The action pattern and reaction mechanism of the endo-1,4-beta-xylanase of the yeast Cryptococcus albidus were investigated using reducing-end (1-3H)-labelled and uniformly 14C-labelled beta-1,4-xylooligosaccharides up to xylopentaose. The enzyme was found to catalyze degradation of oligosaccharides also by other pathways than a simple hydrolytic ...

Two different endo-1,4-beta-xylanases, designated XA-1 and XA-2, and one beta-xylosidase (XD-1) have been purified by column chromatography to apparent homogeneity from the extracellular culture fluid of Phlebia radiata grown on wheat bran. The molecular masses of XA- 1, XA-2 and XD-1 were 18.6, 15.8 kDa, and 27 kDa, respectively.

This experiment was carried out to evaluate the effect of endo-1,4-beta-xylanase with traces of endo-1,4-beta-glucanase on the performance of 240 piglets (Landrace × Yorkshire × Duroc) with an initial body (BW) of 5.90 ± 0.6 kg. The mixed sex piglets were randomly assigned to 3 treatments with 10 replicates per treatment, and 8 heads per replicate pen.

The active site of an acidic endo-1,4-β-xylanase of Aspergillus niger
Biochimica et Biophysica Acta (BBA) - Protein Structure and Molecular Enzymology, 1983The substrate binding site of an acidic endo-1,4-beta-xylanase (1,4-beta-D-xylan xylanohydrolase, EC 3.2.1.8) of Aspergillus niger was investigated using 1,4-beta-xylooligosaccharides (1-3H)-labelled at the reducing end. Bond cleavage frequencies and V/Km parameters of the oligosaccharides were determined under conditions of unimolecular hydrolysis and,

Functional identification of the cDNA coding for a wheat endo-1,4-beta-D-xylanase inhibitor.
FEBS letters, 2002Using expressed sequence tag data, we obtained a full-length cDNA encoding a wheat protein inhibitor of xylanases (XIP-I). The 822 bp open reading frame encoded a protein of 274 amino acids with a molecular mass of 30.2 kDa, in excellent agreement with the native protein.

An endo-1,4-β-xylanase-encoding gene from Agaricus bisporus is regulated by compost-specific factors
Journal of Molecular Biology, 1998Compost is the preferred substrate for growth of the edible fungus Agaricus bisporus. Utilization of compost requires the production of enzymes involved in degradation of lignocellulolytic components. For molecular characterization of these processes we are isolating the encoding genes. By applying heterologous screening techniques, we have cloned such
