Dual Targeting of Tau Kinases and Autophagy by Abemaciclib Independent of CDK4/6 Inhibition
Abemaciclib rescues cognitive function and attenuates tau pathology in Alzheimer's disease models, but not through its known cancer‐fighting mechanism. This study demonstrates that abemaciclib operates independently of CDK4/6 inhibition, instead directly targeting tau kinases CaMKII and GSK3β while simultaneously promoting autophagic clearance of ...

Upregulated TRMT6 forms aberrant hypermethylation of a specific tRNA pool and serves as a predictor of poor prognosis in TNBC. This m1A modification in tRNAs enhances translation of FTH1 and FTL, reducing the pool of bioavailable Fe2⁺. Reduced Fe2+ availability impairs RSL3‐induced lipid peroxidation and tumor progression.

ERM Inhibition Confers Ferroptosis Resistance through ROS‐Induced NRF2 Signaling
ERM inhibition disrupts ERM‐actin interactions, elevating ROS and triggering KEAP1 degradation, which stabilizes and activates NRF2. Nuclear NRF2 induces cytoprotective genes, notably HMOX1, enhancing redox buffering and suppressing lipid peroxidation to resist erastin‐induced ferroptosis.

Single‐Mitochondrion ATP Profiling Directs Discovery of Targetable OXPHOS Dependency in Cancers
MitoATP‐nFCM integrates nano‐flow cytometry with fluorogenic probes (ATP/membrane potential) and antibodies to quantify mitochondrial metabolites and protein expression at single‐organelle resolution, exposing oxidative phosphorylation (OXPHOS)‐driven metabolic rewiring in cancers.

This study reveals that XBP1s drives production of circTspan3, a circular RNA that strengthens cartilage by boosting anabolic activity and limiting cell death. Phosphorylated ANXA2 directs circTspan3 into exosomes, enabling paracrine repair. Exosomal circTspan3 expands growth‐plate cartilage and promotes in vivo regeneration, highlighting its promise ...

The endocrine system is interested by several autoimmune diseases, characterized by different impact and severity, according to the organs involved. Autoimmune thyroid disorders (i.e. Hashimoto's thyroiditis and Graves' disease) and type 1 diabetes mellitus are the most common autoimmune endocrine disorders, while hypophysitis, adrenalitis (90% of ...

Endocrine diseases are rare and can present very subtly in the neonatal period. Most are diagnosed using newborn screening in the United States; however, some infants may present with false negatives or more subtle findings. Endocrine etiologies should be considered during the management of critically ill infants.
