RNase Y is a key endoribonuclease affecting global mRNA stability in Bacillus subtilis. Its characterization provided the first evidence that endonucleolytic cleavage plays a major role in the mRNA metabolism of this organism.

Significance Statement A large‐scale mutational analysis revealed that introducing a single G–C base pair upstream of the first DCL1 cleavage site in shc precursors enhances artificial microRNA accumulation and silencing efficiency, providing an optimized amiRNA precursor for improved gene regulation and biotechnological applications.
