In the experiments reported here we monitored circulatory changes during endotoxemia and their relation to portal and systemic levels of endothelin-1 (ET-1). Piglets were monitored cardiovascularly under ketamine anesthesia for 6 h after an endotoxin infusion (saline controls).

Intracerebroventricular (i.c.v.) or intrathecal (i.t.) administration of morphine to mice antagonized the abdominal constriction induced by an i.p. injection of endothelin-1 (ET-1; 0.1 mg/kg). The ED50 values (95% confidence intervals) were 39.3 (16.5-80.2) ng and 1.5 (0.8-4.9) ng, respectively.
