Symmetrical 2,7‐disubstituted 9H‐fluoren‐9‐one as a novel and promising scaffold for selective targeting of SIRT2

Lead optimization was conducted and a new scaffold of SIRT2‐selective inhibitors was discovered. The top compound SG3 selectively inhibited SIRT2 with an IC50 value of 1.95 μ M and reduced the MCF‐7 cell viability, accompanied by hyperacetylation of α‐tubulin. In silico studies verified the binding ability of SG3 to SIRT2.
