Phosphorylation inhibits guanine nucleotide exchange on eukaryotic initiation factor 2

Nature, 1982
There is extensive evidence that protein synthesis in reticulocyte lysates is inhibited during haem deficiency because of phosphorylation of initiation factor eIF-2 by a protein kinase (the haem-controlled represser, HCR)1–5. Other protein kinases, which are activated by low concentrations of double-stranded RNA, have also been identified in ...

Characteristics of the eukaryotic initiation factor 2 associated 67-kDa polypeptide

Biochemistry, 1993
A eukaryotic initiation factor 2 (eIF-2) associated 67-kDa polypeptide (p67) protects the eIF-2 alpha-subunit from eIF-2 kinase(s) catalyzed phosphorylation, and this promotes protein synthesis in the presence of active eIF-2 kinase(s), [Datta, B., et al. (1988) Proc. Natl. Acad. Sci. U.S.A. 85, 3324-3328].

The nature of the interaction of eukaryotic initiation factor 2 with double‐stranded RNA

European Journal of Biochemistry, 1984
In addition to binding messenger RNA molecules at specific sequences, eukaryotic initiation factor 2 (eIF‐2) also binds to double‐stranded RNA (dsRNA). The dsRNA is a powerful inhibitor of initiation of eukaryotik translation, causing the inactivation of eIF‐2, but in the presence of certain mRNA templates, dsRNA fails to establish inhibition.

Eukaryotic type translation initiation factor 2: Structure–functional aspects

Biochemistry (Moscow), 2011
Translation initiation factor 2 (IF2) is one of key components of the translation initiation system in living cells. In bacteria IF2 is a multidomain monomeric protein, while in eukaryotic and archaean cells e/aIF2 is heterotrimer (αβγ). Data, including our own, on eukaryotic type translation initiation factor 2 (e/aIF2) structure and functioning are ...

Roles of Eukaryotic Initiation Factor 2 Ancillary Factors in the Regulation of Eukaryotic Protein Synthesis Initiation

1982
Publisher Summary This chapter discusses a possible mechanism of protein synthesis inhibition by eukaryotic initiation factor-2 (eIF-2) kinases, heme-regulated protein synthesis inhibitor, and double-stranded RNA-activated inhibitor; it discusses the roles of four eIF-2 ancillary protein factors in the complex regulation of protein synthesis ...

Binding of ATP and messenger RNA by the β-subunit of eukaryotic initiation factor 2

Biochimie, 1992
In addition to forming a ternary complex with Met-tRNA(f) and GTP, eukaryotic initiation factor 2 (eIF-2) recognizes a specific site in mRNA molecules. Both binding activities are regulated by ATP, which itself binds tightly and specifically to eIF-2.

Eukaryotic translation initiation factor 2 α phosphorylation as a therapeutic target in diabetes

Expert Review of Endocrinology & Metabolism, 2014
Regulation of mRNA translation is of vital importance for a cell to adapt to environmental changes. To serve this purpose, intricate mechanisms controlling mRNA translation have evolved, of which the eukaryotic initiation factors eIF2 and eIF4 represent essential regulatory nodes for both stress sensing and signal transduction. Stress sensing by eIF2 α

Suppression of osteoclastogenesis through phosphorylation of eukaryotic translation initiation factor 2 alpha

Journal of Bone and Mineral Metabolism, 2013
In response to various stresses including viral infection, nutrient deprivation, and stress to the endoplasmic reticulum, eukaryotic translation initiation factor 2 alpha (eIF2α) is phosphorylated to cope with stress induced apoptosis. Although bone cells are sensitive to environmental stresses that alter the phosphorylation level of eIF2α, little is ...

Quantifying the Binding of Fluorescently Labeled Guanine Nucleotides and Initiator tRNA to Eukaryotic Translation Initiation Factor 2

2022
The translation initiation factor eIF2 is critical for protein synthesis initiation, and its regulation is central to the integrated stress response (ISR). eIF2 is a G protein, and the activity is regulated by its GDP or GTP-binding status, such that only GTP-bound eIF2 has high affinity for initiator methionyl tRNA.

Mutually exclusive binding of messenger RNA and initiator methionyl transfer RNA to eukaryotic initiation factor 2

Biochemical and Biophysical Research Communications, 1979
Abstract Eukaryotic initiation factor 2 (eIF-2), purified to at least 98% homogeneity as judged by polyacrylamide gel electrophoresis in sodium dodecyl sulfate, and containing no detectable amounts of eukaryotic initiation factor 4B (eIF-4B), is active both in the binding of Met-tRNA and in the binding of globin mRNA.
