An acid‐cleavable Tf‐modified engineering exosome‐based brain‐targeting delivery system (ACTE) was proposed using metabolic glycoengineering and bioorthogonal click chemistry, enabling co‐delivery of siTGF‐β and DOX for GBM treatment. ACTE was featured by higher BBB transcytosis efficiency and hierarchical targeting delivery to GBM cells.

Synthetic Biology‐Based Engineering Cells for Drug Delivery
Synthetic biology‐based therapeutic cells for drug delivery: Engineered cells effectively transport and release drugs through their intelligence and controllability, such as feedback pathways and external stimuli (e.g., light and ultrasound). The integration of nanotechnology, including nanoparticles, biofilms, and artificial cells, enhances ...

Engineered Exosome‐Based Senolytic Therapy Alleviates Stroke by Targeting p21+CD86+ Microglia
We identified a harmful immune cell type, p21+CD86+ microglia, driving post‐stroke inflammation and developed a targeted exosome‐based therapy (Que@micro‐Exo) to eliminate these cells. This innovative treatment reduced inflammation, alleviated brain damage, and improved recovery in stroke models, offering a promising strategy for stroke and other ...
