Pre‐analytical optimization of cell‐free DNA and extracellular vesicle‐derived DNA for mutation detection in liquid biopsies

open access: yesMolecular Oncology, EarlyView.
Pre‐analytical handling critically determines liquid biopsy performance. This study defines practical best‐practice conditions for cell‐free DNA (cfDNA) and extracellular vesicle–derived DNA (evDNA), showing how processing time, storage conditions, tube type, and plasma input volume affect DNA integrity and mutation detection.

Interrogating the immune landscape of microsatellite stable RAS‐mutated colon cancer

open access: yesMolecular Oncology, EarlyView.
COLOSSUS project RAS‐mutated MSS colon cancer study explored transcriptomics and immune cell density by immunohistochemistry (IHC), Immunoscore (IS), ISIC/TuLIS scores, mutation counts, and detected different prevalences but similar microenvironment composition across immune markers with clinical relevance for future immunotherapy combination ...

Metastasis on pause: How dormant tumor cells stay hidden within the tumor microenvironment and evade immune surveillance

open access: yesMolecular Oncology, EarlyView.
Dormant cancer cells can hide in distant organs for years, evading treatment and the immune system. This review highlights how signals from the surrounding tissue and immune environment keep these cells inactive or trigger their reawakening. Understanding these mechanisms may help develop therapies to eliminate or control dormant cells and prevent ...

IAPSO Standard Seawater: definition of the uncertainty in the calibration procedure, and stability of recent batches [PDF]

open access: yes, 2007
Standard seawater (SSW) has been employed by oceanographers as a reference material in the determinationof salinity for over a century. In all that time, this is the first study to determine the uncertaintyof the SSW manufacturing process.

Evaluation of expanded uncertainties in luminous intensity and illuminance calibrations

open access: yesApplied Optics, 2008
Detector-based calibrating methods and expressions for calculation of photometric uncertainties related to uncertainties in the calibrations of luminous intensity of a light source, illuminance responsivity of a photometer head, and calibration factors of an illuminance meter are discussed.

Finding novel vulnerabilities of hypomorphic BRCA1 alleles

open access: yesMolecular Oncology, EarlyView.
Synthetic lethality screens performed to identify novel vulnerabilities often model complete gene loss, thereby overlooking patient‐derived hypomorphic mutations. In this study, we have performed genome‐wide CRISPR screens on BRCA1 hypomorphic mutations, showing BRCA1I26A behaves like wild‐type, while BRCA1R1699Q mimics deficiency. Furthermore, we have

A Proposed Estimation of the Expanded Uncertainty of Charpy Impact Testers

open access: yesAsian Journal of Applied Sciences, 2020
Uncertainty estimation is one of the very delicate tasks in the field of measurements. For the purpose of calibration of Charpy impact testing machines, it is necessary to evaluate and identify the expanded uncertainty. Factors affecting the uncertainty estimations are; the uncertainty of reference force and length measuring devices and its long-term ...

Screening and epitope characterization of Nidogen‐2‐specific nanobodies

open access: yesFEBS Open Bio, EarlyView.
Camel immunization and phage display were employed to generate high‐affinity VHH nanobodies against Nidogen‐2. After library construction, biopanning, ELISA screening, sequencing, and recombinant expression, selected nanobodies were purified and characterized, leading to the preliminary exploration of a nanobody‐based sandwich ELISA for specific ...

The skills required for transition to university and study in biological sciences: A student perspective

open access: yesFEBS Open Bio, EarlyView.
Bioscience students were asked for their opinions on the value and teaching of skills. 204 responded that teamwork, time management and study skills are necessary to reach University, that scientific writing, research, laboratory and presentation skills are taught effectively during their studies, while other skills are gained inherently through study ...
