Genetic variants in ELOVL2 and HSD17B12 predict melanoma‐specific survival [PDF]

open access: yes, 2019
Fatty acids play a key role in cellular bioenergetics, membrane biosynthesis and intracellular signaling processes and thus may be involved in cancer development and progression.

An unexpected link between fatty acid synthase and cholesterol synthesis in proinflammatory macrophage activation

open access: yesJournal of Biological Chemistry, 2018
Different immune activation states require distinct metabolic features and activities in immune cells. For instance, inhibition of fatty acid synthase (FASN), which catalyzes the synthesis of long-chain fatty acids, prevents the proinflammatory response ...

CDK11 inhibition induces cytoplasmic p21WAF1 splice variant by p53 stabilisation and SF3B1 inactivation

open access: yesMolecular Oncology, EarlyView.
CDK11 inhibition stabilises the tumour suppressor p53 and triggers the production of an alternative p21WAF1 splice variant p21L, through the inactivation of the spliceosomal protein SF3B1. Unlike the canonical p21WAF1 protein, p21L is localised in the cytoplasm and has reduced cell cycle‐blocking activity.

Resveratrol Modulates Desaturase Expression and Fatty Acid Composition of Cultured Hepatocytes

open access: yesFrontiers in Nutrition, 2018
This study aimed to evaluate whether resveratrol (RSV) and its microbial metabolites dihydro-resveratrol (DHR) and lunularin (LUN) affected fatty acid metabolism and omega-3 polyunsaturated fatty acid (n3-PUFA) synthesis in cultured hepatocytes.

Prenatal hyperandrogenism induces alterations that affect liver lipid metabolism [PDF]

open access: yes, 2016
Prenatal hyperandrogenism is hypothesized as one of the main factors contributing to26 the development of polycystic ovary syndrome (PCOS). PCOS patients have high risk27 of developing fatty liver and steatosis.

ATF4‐mediated stress response as a therapeutic vulnerability in chordoma

open access: yesMolecular Oncology, EarlyView.
We screened 5 chordoma cell lines against 100+ inhibitors of epigenetic and metabolic pathways and kinases and identified halofuginone, a tRNA synthetase inhibitor. Mechanistically halofuginone induces an integrated stress response, with eIF2alpha phosphorylation, activation of ATF4 and its target genes CHOP, ASNS, INHBE leading to cell death ...

Mechanisms by which tumor necrosis factor stimulates hepatic fatty acid synthesis in vivo

open access: yesJournal of Lipid Research, 1988
We have previously shown that bolus intravenous administration of tumor necrosis factor (TNF) to normal rats results in a rapid (within 90 min) stimulation of hepatic fatty acid synthesis, which is sustained for 17 hr.

Circadian clock and lipid metabolism disorders: a potential therapeutic strategy for cancer

open access: yesFrontiers in Endocrinology, 2023
Recent research has emphasized the interaction between the circadian clock and lipid metabolism, particularly in relation to tumors. This review aims to explore how the circadian clock regulates lipid metabolism and its impact on carcinogenesis ...

Tumour–host interactions in Drosophila: mechanisms in the tumour micro‐ and macroenvironment

open access: yesMolecular Oncology, EarlyView.
This review examines how tumour–host crosstalk takes place at multiple levels of biological organisation, from local cell competition and immune crosstalk to organism‐wide metabolic and physiological collapse. Here, we integrate findings from Drosophila melanogaster studies that reveal conserved mechanisms through which tumours hijack host systems to ...

Engineering of fatty acid production and secretion in Saccharomyces cerevisiae [PDF]

open access: yes, 2009
Production of renewable liquid biofuels that can substitute fossil fuel, has emerged as a major challenge for applied biology. Biodiesel, in the form of fatty acid esters, produced by oleaginous microorganisms could be an attractive alternative, since ...
