Clinical Features of Cellular Senescence Pathways in Severe Asthma
Allergy, Volume 81, Issue 9, Page 3273-3285, September 2026.In bronchial biopsies, SASP and p53 pathway enrichment scores are elevated in severe asthma versus non‐severe asthma and healthy controls. SASP enrichment is validated in the independent NOVA cohort. SASP enrichment is also elevated in nasal brushings of participants with nasal polyps, independently of asthma status.Woo‐Jung Song, Nazanin Zounemat Kermani, Ali Versi, Stephany Sánchez‐Ovando, Jodie Louise Simpson, Peter A. Allergy, Volume 81, Issue 9, Page 3286-3299, September 2026.Longer asthma duration is associated with increased airway non‐T2 inflammation and reduced airway and systemic T2 inflammation. Sputum CXCL9 is independently associated with duration and may predict a reduced poor response to anti‐IL‐4Rα therapy. Airway sampling and combined T2 and non‐T2 targeted strategies may be needed in long‐standing asthma. CXCL9,Freda Yang, Sujin Seo, Takehiro Hasegawa, Chloe I. Allergy, Volume 81, Issue 9, Page 3300-3317, September 2026.This study distinguishes severe asthma–related airway transcriptomic changes from the effects of high‐dose ICS. Most dysregulated genes in severe asthma show concordance with corticosteroid responses. Corticosteroid‐resistant T2‐high asthma featured T2‐high and epithelial barrier gene upregulation, while T2‐low asthma featured Th1/IL‐17 signatures. Airway remodeling is increasingly recognized as a major determinant of asthma progression, fixed airflow limitation, and long‐term morbidity, particularly in severe disease. Although biologic therapies have transformed outcomes by reducing exacerbations and systemic corticosteroid exposure, their potential to modify structural airway ...Sebastiano Gangemi, Sara Manti, Johann Christian Virchow, Giorgio Walter Canonica +3 morewiley +1 more sourceAgonist‐specific conformational dynamics at the β2‐adrenoceptor dictate allosteric modulation of Gαs signalling and bronchodilation

β2‐adrenoceptor (β2AR) agonists are the cornerstone of asthma therapy and promote bronchodilation through Gαs signalling in airway smooth muscle (ASM), but their efficacy is limited by β‐arrestin‐mediated β2AR desensitization.Sushrut D. The landscape of asthma management is undergoing significant transformation. This change is driven by several factors: deeper understanding of asthma pathophysiology, appreciation of the long‐term harm of oral corticosteroids, biological treatments that dramatically improve patient outcomes with asthma remission becoming a realistic goal, and ...John Politis, Kerry Hancock, Jo A Douglass, Paul Leong, Philip Bardin +4 morewiley +1 more source
