A General Workflow for Tangential Flow Filtration Perfusion Scale‐Up

open access: yesBiotechnology and Bioengineering, EarlyView.
ABSTRACT Tangential flow filtration (TFF) perfusion using hollow‐fiber modules offers substantial advantages over other mammalian cell retention technologies. Recently, increased awareness of Starling flow has helped to enhance filtration performance to similar, or even higher levels compared to alternative technologies.

Purification of Disassembled Capsid Protein by Hydroxyapatite Chromatography Enhances Virus‐Like Particle Recovery

open access: yesBiotechnology and Bioengineering, EarlyView.
ABSTRACT Virus‐like particles (VLPs) are a promising modality with extensive applications in prophylactic and therapeutic vaccine design. Human Papillomavirus (HPV) vaccines are one such example where VLP vaccine applications have led to the successful reduction in HPV‐associated diseases, such as cervical cancer.

Combinatorial Metabolic Engineering of Alkane Biosynthesis in the Osmotolerant Yeast Debaryomyces hansenii CBS 767

open access: yesBiotechnology and Bioengineering, EarlyView.
ABSTRACT Debaryomyces hansenii is a yeast with diverse potential applications like producing oleochemicals from biomass. Yet, combinatorial metabolic engineering in D. hansenii is limited because, like many nonconventional organisms, key genomic data and genetic elements are missing. Here, we report phenotypic characterization, genomic integration loci,

Whole‐Cell Production of Resolvin E2 and 17R‐Resolvin D5 by a Double‐Oxygenating 15S‐Lipoxygenase With 5S‐Lipoxygenase Activity From Pseudobdellovibrionaceae bacterium

open access: yesBiotechnology and Bioengineering, EarlyView.
A recombinant whole‐cell platform expressing a bacterial 15S‐LOX exhibiting 5S‐LOX activity enabled the stereoselective production of RvE2 and 17R‐RvD5 from 18R‐HEPE and 17R‐HDHA, achieving final concentrations of 0.57 mM and 1.48 mM from 4.0 mM substrates, respectively, under optimized conditions.

Characterization of Immune Responses to RBD‐SD1 Nanoparticle Vaccines Derived From ACE2‐Capable HKU5‐CoV‐2, NeoCoV, and HKU5 Merbecoviruses

open access: yesBiotechnology and Bioengineering, EarlyView.
SpyCatcher‐mi3 nanoparticles presenting HKU5, NeoCoV, and HKU5‐CoV‐2 RBD‐SD1 induced strong, cross‐reactive IgG responses in immunized mice. The surrogate virus neutralization test demonstrated strong activity in sera from homologous vaccination. ABSTRACT Recent discoveries of merbecoviruses such as HKU5, NeoCoV, and HKU5‐CoV‐2 that use angiotensin ...
