This study introduces a new and simple method for the determination of free fluorescein in plasma on the basis of determined total free plasma fluorescense and the free fraction of fluorescence.

Fluorescein angiography (FA) is an imaging modality used to study the circulation of the retina and choroid. About 5 mL of 10% sterile aqueous solution of fluorescein dye (or 2-3 mL of a 25% solution) is injected in the antecubital vein. The fluorescein molecules excited by a blue light (465-490 nm) emit green-yellow light (520-530 nm) (Fig. 2.1).

To evaluate patency of the lacrimal system, fluorescein was instilled in the conjunctival cul-de-sac and the posterior oropharynx was examined with ultraviolet light. In 20 normal patients fluorescein was evident in one to 30 minutes. In cases of complete nasolacrimal duct blockage no fluorescein appeared after two hours.
