Detecting and Predicting Forecast Breakdowns [PDF]
We propose a theoretical framework for assessing whether a forecast model estimated over one period can provide good forecasts over a subsequent period.

Diversity and complexity in neural organoids
Neural organoid research aims to expand genetic diversity on one side and increase tissue complexity on the other. Chimeroids integrate multiple donor genomes within single organoids. Self‐organising multi‐identity organoids, exogenous cell seeding, or enforced assembly of region‐specific organoids contribute to tissue complexity.

A General Forecast-error Taxonomy [PDF]
The paper considers the sources of forecast errors and their consequences in an evolving economy subject to structural breaks, forecasting from mis-specified, data-based models. A model-free taxonomy of forecast errors highlights that deterministic shifts

Hyperosmotic stress induces PARP1‐mediated HPF1‐dependent mono(ADP‐ribosyl)ation
Sorbitol‐induced hyperosmotic stress rapidly induces reversible mono(ADP‐ribosyl)ation (MARylation) on PARP1 without the signs of genotoxic signaling. We show that PARP1 autoMARylation is HPF1 dependent and forms hydroxylamine‐resistant O‐glycosidic linkages.

An isoform of 14‐3‐3 protein regulates transbilayer lipid movement at the plasma membrane
Loss of 14‐3‐3ζ in CHO cells confers resistance to exogenous phosphatidylserine (PS) and impairs endocytosis‐independent inward flip‐flop of fluorescent PS at the plasma membrane. RNAi‐mediated knockdown reproduces this defect, while no additive effect is seen in ATP11C‐deficient cells.

The ubiquitin ligase RNF115 is required for the clearance of damaged lysosomes
Upon lysosomal rupture, an E3 ubiquitin ligase RNF115 translocates from the cytosol to the damaged lysosomal membrane. Moreover, RNF115 depletion impairs the clearance of damaged lysosomes, identifying it as a key regulator of lysosomal quality control.

It starts in October, the merciless grey morass that gets worse in November and sucks you down into a boghole of misery. It's deep, and oozing, nearly as deep as the silence of a middle aged man in a McGahern novel. Rain. Imagine what all that dreck and drear does to the psyche. It's like the inside of The Poor Mouth and The Hard Life in there.

pH‐mediated activation of the lysosomal arginine sensor SLC38A9
Cells monitor nutrient levels via the lysosomal transporter SLC38A9 to activate the mechanistic target of rapamycin complex 1 (mTORC1). This study reveals that SLC38A9 function is regulated by pH. We identified histidine 544 as a critical pH sensor that undergoes conformational changes to control amino acid efflux from lysosomes; therefore, it ...
