C2α‐carbanion‐protonating glutamate discloses tradeoffs between substrate accommodation and reaction rate in actinobacterial 2‐hydroxyacyl‐CoA lyase

open access: yesFEBS Open Bio, EarlyView.
Enzymes of the 2‐hydroxyacyl‐CoA lyase group catalyze the condensation of formyl‐CoA with aldehydes or ketones. Thus, by structural adaptation of active sites, practically any pharmaceutically and industrially important 2‐hydroxyacid could be biotechnologically synthesized. Combining crystal structure analysis, active site mutations and kinetic assays,

Cointegration and Forward and Spot Exchange Rate Regressions [PDF]

open access: yes
In this paper we investigate in detail the relationship between models of cointegration between the current spot exchange rate, st, and the current forward rate, ft, and models of cointegration between the future spot rate, st+1, and ft and the ...

PARP inhibitors induce a senescence phenotype in non‐small cell lung carcinoma cell lines

open access: yesFEBS Open Bio, EarlyView.
Talazoparib is the most potent inducer of senescence among different PARP1 inhibitors in human NSCLC cells. In the absence of PARP, no senescence phenotype was observed, demonstrating that PARP1 is necessary for the induction of senescence by this inhibitor.

Forward Looking and Myopic Regional Computable General Equilibrium Models. How Significant is the Distinction? [PDF]

open access: yes
We present a stylized intertemporal forward-looking model able that accommodates key regional economic features, an area where the literature is not well developed.

Proteasomal degradation of intracellularly expressed Amblyomin‐X limits suicide gene therapy potential in melanoma cells

open access: yesFEBS Open Bio, EarlyView.
This study explores the feasibility of expressing the antitumoral protein Amblyomin‐X through a suicide gene therapy approach and investigates its intracellular fate after gene delivery. Although the gene is efficiently expressed, melanoma cells rapidly degrade the Amblyomin‐X protein via proteasome activity.

Combined 5‐aminolevulinic acid and ferric ammonium citrate treatment promotes hair follicle growth by activating dermal papilla cells

open access: yesFEBS Open Bio, EarlyView.
5‐Aminolevulinic acid combined with ferric ammonium citrate (5‐ALA/FAC) stimulates dermal papilla cell activity and promotes hair follicle growth. The treatment enhances ERK and AKT signaling, increases hair‐inductive gene expression, and restores dermal papilla function suppressed by dihydrotestosterone and oxidative stress, resulting in enhanced hair

Finite dimensional Markovian realizations for stochastic volatility forward rate models [PDF]

open access: yes
We consider forward rate rate models of HJM type, as well as more general infinite dimensional SDEs, where the volatility/diffusion term is stochastic in the sense of being driven by a separate hidden Markov process.

Evolutionarily divergent DUF4465 domains have a common vitamin B12‐binding function

open access: yesFEBS Open Bio, EarlyView.
We show that DUF4465 family proteins, widespread across bacteria from gut microbiomes, hydrothermal vents, and soil, share a common vitamin B12‐binding function. These augmented β‐jellyroll proteins bind vitamin B12 via extended loops. Our findings establish sequence‐diverse DUF4465 proteins as a widespread class of B12‐binding proteins, highlighting ...

Replacing Backpropagation with the Forward-Forward (FF) Algorithm in Transformer Models: A Theoretical and Empirical Study on Scalable and Efficient Gradient-Free Training

open access: yesTehnički Glasnik
This study proposes a novel integration of the Forward-Forward (FF) algorithm into Transformer architectures as an efficient and gradient-free alternative to Backpropagation (BP). Motivated by the computational limitations of BP-such as high memory usage
