Single‐Cell RNA Editing Identifies T Cell ADAR1 as a Key Regulator of Immune Exhaustion and Anti‐PD‐1 Resistance in Colorectal Cancer

open access: yesAdvanced Science, EarlyView.
Single‐cell RNA editing analysis identifies ADAR1 as a regulator of dysfunctional T cell states in colorectal cancer. Elevated ADAR1 activity promotes T cell exhaustion and impairs antitumor immunity partly through TGF‐β‐SMAD signaling, contributing to anti‐PD‐1 resistance and highlighting T cell ADAR1 as a potential therapeutic target and biomarker ...

Mechanism of Action of Botulinum Toxin A in Treatment of Functional Urological Disorders

open access: yesToxins, 2020
Intravesical botulinum toxin (BoNT) injection is effective in reducing urgency and urinary incontinence. It temporarily inhibits the detrusor muscle contraction by blocking the release of acetylcholine (Ach) from the preganglionic and postganglionic ...

An Integrative Strategy Delineates Modular Metabolic Remodeling and Potential Therapeutic Targets Across Metabolic Diseases

open access: yesAdvanced Science, EarlyView.
An integrative single‐cell atlas across multiple metabolic diseases reveals coordinated metabolic modules and disease‐shared versus disease‐specific pathway activities. By systematically comparing scoring strategies, a robust RankAve framework is established. Coupled with network analysis and drug‐target prediction, this resource uncovers cross‐disease

DHODH Drives Sunitinib Resistance Via a Non‐Enzymatic Mechanism by Inhibiting TRIM28 Ubiquitination and Consequent VEGFA Activation in RCC

open access: yesAdvanced Science, EarlyView.
This non‑enzymatic function of DHODH drives sunitinib resistance by competing with TRIM37 to block TRIM28 ubiquitination, thereby stabilizing TRIM28 and activating VEGFA transcription. Disrupting the DHODH–TRIM28 interaction with lisaftoclax restores drug sensitivity.

Targeted Delivery of Indole‐3‐Pyruvic Acid Suppresses Macrophage Ferroptosis to Enhance CD8+ T Cell‐Mediated Immunotherapy Response in Bladder Cancer

open access: yesAdvanced Science, EarlyView.
A microbiota‐derived metabolite, indole‐3‐pyruvic acid, suppresses macrophage ferroptosis through the AHR–NF‐κB–SLC7A11 axis. This preserves CD8+ T cell function in bladder cancer. Macrophage‐targeted nanoparticles enhance indole‐3‐pyruvic acid delivery and overcome resistance to PD‐1 blockade.

Can We Go beyond Pathology? The Prognostic Role of Risk Scoring Tools for Cancer-Specific Survival of Patients with Bladder Cancer Undergoing Radical Cystectomy

open access: yesBiomedicines
Radical cystectomy (RC) remains a mainstay surgical treatment for non-metastatic muscle-invasive and BCG-unresponsive bladder cancer. Various perioperative scoring tools assess comorbidity burden, complication risks, and cancer-specific mortality (CSM ...
