Exploiting metabolic adaptations to overcome dabrafenib treatment resistance in melanoma cells

open access: yesMolecular Oncology, EarlyView.
We show that dabrafenib‐resistant melanoma cells undergo mitochondrial remodeling, leading to elevated respiration and ROS production balanced by stronger antioxidant defenses. This altered redox state promotes survival despite mitochondrial damage but renders resistant cells highly vulnerable to ROS‐inducing compounds such as PEITC, highlighting redox

LINC01116, a hypoxia‐lncRNA marker of pathological lymphangiogenesis and poor prognosis in lung adenocarcinoma

open access: yesMolecular Oncology, EarlyView.
The LINC01116 long noncoding RNA is induced by hypoxia and associated with poor prognosis and high recurrence rates in two cohorts of lung adenocarcinoma patients. Here, we demonstrate that besides its expression in cancer cells, LINC01116 is markedly expressed in lymphatic endothelial cells of the tumor stroma in which it participates in hypoxia ...

Vibration and Buckling Analysis of Functionally Graded Plates Using New Eight-Unknown Higher Order Shear Deformation Theory

open access: yesLatin American Journal of Solids and Structures
In this paper a new eight-unknown higher order shear deformation theory is proposed to study the buckling and free vibration of functionally graded (FG) material plates.

ATF4‐mediated stress response as a therapeutic vulnerability in chordoma

open access: yesMolecular Oncology, EarlyView.
We screened 5 chordoma cell lines against 100+ inhibitors of epigenetic and metabolic pathways and kinases and identified halofuginone, a tRNA synthetase inhibitor. Mechanistically halofuginone induces an integrated stress response, with eIF2alpha phosphorylation, activation of ATF4 and its target genes CHOP, ASNS, INHBE leading to cell death ...

Exact solution for bending analysis of functionally graded micro-plates based on strain gradient theory

open access: yesScience and Engineering of Composite Materials, 2018
In the present article, static analysis of thin functionally graded micro-plates, based on Kirchhoff plate theory, is investigated. Utilizing the strain gradient theory and principle of minimum total potential energy, governing equations of rectangular ...

Analysis of Stability and Vibrations of Porous Power and Sigmoid Functionally Graded Sandwich Plates by the R-Functions Method

open access: yesJournal of Mechanical Engineering, 2023
In this paper, the R-functions method is used for the first time to study the stability and vibrations of porous functionally graded (FG) sandwich plates with a complex geometric shape.

Structural Integrity of Functionally Graded Composite Structure using Mindlin-Type Finite Elements. [PDF]

open access: yes, 2008
In this paper, two new Mindlin-type plate bending elements have been derived for the modelling of functionally graded plate subjected to various loading conditions such as tensile loading, in-plane bending and out-of-plane bending.

Cell surface interactome analysis identifies TSPAN4 as a negative regulator of PD‐L1 in melanoma

open access: yesMolecular Oncology, EarlyView.
Using cell surface proximity biotinylation, we identified tetraspanin TSPAN4 within the PD‐L1 interactome of melanoma cells. TSPAN4 negatively regulates PD‐L1 expression and lateral mobility by limiting its interaction with CMTM6 and promoting PD‐L1 degradation.

PARP inhibition and pharmacological ascorbate demonstrate synergy in castration‐resistant prostate cancer

open access: yesMolecular Oncology, EarlyView.
Pharmacologic ascorbate (vitamin C) increases ROS, disrupts cellular metabolism, and induces DNA damage in CRPC cells. These effects sensitize tumors to PARP inhibition, producing synergistic growth suppression with olaparib in vitro and significantly delayed tumor progression in vivo. Pyruvate rescue confirms ROS‐dependent activity.

Plecstatin inhibits hepatocellular carcinoma tumorigenesis and invasion through cytolinker plectin

open access: yesMolecular Oncology, EarlyView.
The ruthenium‐based metallodrug plecstatin exerts its anticancer effect in hepatocellular carcinoma (HCC) primarily through selective targeting of plectin. By disrupting plectin‐mediated cytoskeletal organization, plecstatin inhibits anchorage‐dependent growth, cell polarization, and tumor cell dissemination.
