Exploring the role of cyclin D1 in the pathogenesis of multiple myeloma beyond cell cycle regulation

open access: yesMolecular Oncology, EarlyView.
Cyclin D1 overexpression altered the cell adhesion pathway, while cyclin D2 upregulation had less impact on pathway enrichment analysis. Multiple myeloma (MM) patients with cyclin D1 overexpression showed reduced CD56 expression and increased circulating tumor cells (CTC) levels, suggesting that cyclin D1 may contribute to MM cell dissemination ...

β‐TrCP overexpression enhances cisplatin sensitivity by depleting BRCA1

open access: yesMolecular Oncology, EarlyView.
Low levels of β‐TrCP (Panel A) allow the accumulation of BRCA1 and CtIP, which facilitate the repair of cisplatin‐induced DNA damage via homologous recombination (HR) and promote tumor cell survival. In contrast, high β‐TrCP expression (Panel B) leads to BRCA1 and CtIP degradation, impairing HR repair, resulting in persistent DNA damage and apoptosis ...

Divergent on-target off-tumor effects by CAR T and CAR NK cells suggest different efficacy and safety of cell therapies

open access: yesOncoImmunology
CAR-based cell therapies have shown clinical success in treating various cancers, with CAR T cell therapies entering the clinical route and CAR NK cell therapies being evaluated in early-stage clinical trials.

Gene Therapy Evolution: How Gene Therapy has Evolved Preventive Medicine [PDF]

open access: yes, 2014
Gene therapy technologies offers a prospect for preventive medicine; before the trait can harm the recipient, genetically engineered transgenes will either alter the genetic disorder beforehand, or allow for regeneration of the tissue.

The critical role of DNA damage‐inducible transcript 4 (DDIT4) in stemness character of leukemia cells and leukemia initiation

open access: yesMolecular Oncology, EarlyView.
Stemness properties, including quiescence, self‐renewal, and chemoresistance, are closely associated with leukemia relapse. Here, we demonstrate that DNA damage‐inducible transcript 4 (DDIT4) is induced in the hypoxic bone marrow niche and is essential for maintaining the stemness of AML1‐ETO9a leukemia cells.

Liver fibrosis negatively impacts in vivo gene transfer to murine hepatocytes

open access: yesNature Communications
Liver fibrosis occurs in several genetic and acquired disease conditions, leading to alterations of the tissue and metabolism, which may adversely affect viral vector-mediated gene therapy.
