Bridging the gap: Multi‐stakeholder perspectives of molecular diagnostics in oncology

open access: yesMolecular Oncology, EarlyView.
Although molecular diagnostics is transforming cancer care, implementing novel technologies remains challenging. This study identifies unmet needs and technology requirements through a two‐step stakeholder involvement. Liquid biopsies for monitoring applications and predictive biomarker testing emerge as key unmet needs. Technology requirements vary by

Generalized Linear Mixed-Effects Models for the Analysis of Odor Detection Data [PDF]

open access: yes, 2007
Olfactory detection has become a science of interest. Seven individuals’ odor detection abilities are explored and an attempt is made to characterize all subjects with one generalized linear mixed effects model.

Patient‐specific pharmacogenomics demonstrates xCT as predictive therapeutic target in colon cancer with possible implications in tumor connectivity

open access: yesMolecular Oncology, EarlyView.
This study integrates transcriptomic profiling of matched tumor and healthy tissues from 32 colorectal cancer patients with functional validation in patient‐derived organoids, revealing dysregulated metabolic programs driven by overexpressed xCT (SLC7A11) and SLC3A2, identifying an oncogenic cystine/glutamate transporter signature linked to ...

Aggressive prostate cancer is associated with pericyte dysfunction

open access: yesMolecular Oncology, EarlyView.
Tumor‐produced TGF‐β drives pericyte dysfunction in prostate cancer. This dysfunction is characterized by downregulation of some canonical pericyte markers (i.e., DES, CSPG4, and ACTA2) while maintaining the expression of others (i.e., PDGFRB, NOTCH3, and RGS5).

Fast stable direct fitting and smoothness selection for Generalized Additive Models [PDF]

open access: yes, 2007
Existing computationally efficient methods for penalized likelihood GAM fitting employ iterative smoothness selection on working linear models (or working mixed models).

Bayesian inference for generalized linear mixed models [PDF]

open access: yesBiostatistics, 2009
Generalized linear mixed models (GLMMs) continue to grow in popularity due to their ability to directly acknowledge multiple levels of dependency and model different data types. For small sample sizes especially, likelihood-based inference can be unreliable with variance components being particularly difficult to estimate.

Effect of chemotherapy on passenger mutations in metastatic colorectal cancer

open access: yesMolecular Oncology, EarlyView.
Changes in passenger mutation load and predicted immunotherapy response after chemotherapy treatment. Tumor cells rich with passenger mutations have increased sensitivity to chemotherapy. Correlation of passenger mutations with neoantigen load suggests highly mutated clones promote a more effective response to immunotherapy, and therefore, first‐line ...

Modeling dependencies between rating categories and their effects on prediction in a credit risk portfolio [PDF]

open access: yes, 2006
The internal-ratings based Basel II approach increases the need for the development of more realistic default probability models. In this paper we follow the approach taken in McNeil and Wendin (2006) by constructing generalized linear mixed models for ...

Developing evidence‐based, cost‐effective P4 cancer medicine for driving innovation in prevention, therapeutics, patient care and reducing healthcare inequalities

open access: yesMolecular Oncology, EarlyView.
The cancer problem is increasing globally with projections up to the year 2050 showing unfavourable outcomes in terms of incidence and cancer‐related deaths. The main challenges are prevention, improved therapeutics resulting in increased cure rates and enhanced health‐related quality of life.

Genome-Wide Association Study for Nodule Traits in Guar

open access: yesCurrent Issues in Molecular Biology
Guar [Cyamopsis tetragonoloba (L.) Taub] is a diploid legume crop cultivated for galactomannan (guar gum) extracted from the endosperm of the seed. Previous studies have suggested that nodulation of guar can be poor in field conditions; however, solid ...
