Genome mining and prospects for antibiotic discovery

Current Opinion in Microbiology, 2019
Natural products are a rich source of bioactive compounds that have been used successfully in the areas of human health from infectious disease to cancer; however, traditional fermentation-based screening has provided diminishing returns over the last 20-30 years.

Genome mining and in vivo characterization of triceptides

Triceptides are cyclophane-containing ribosomally synthesized and post-translationally modified peptides (RiPPs) produced by bacteria. The class defining three-residue cyclophane is installed post-translationally by radical SAM enzymes (triceptide synthases) between the aromatic ring of Ω1 and the Cβ of X3 on Ω1-X2-X3 motifs.

Genome mining for new enediyne antibiotics

Current Opinion in Chemical Biology
Enediyne antibiotics epitomize nature's chemical creativity. They contain intricate molecular architectures that are coupled with potent biological activities involving double-stranded DNA scission. The recent explosion in microbial genome sequences has revealed a large reservoir of novel enediynes. However, while hundreds of enediyne biosynthetic gene
