Clinical application of GnRH-antagonists
Molecular and Cellular Endocrinology, 2000Due to the different pharmacological mode of action, GnRH-antagonists seem to open up new avenues to hormonal treatment in several indications. Although it may be still too early to speculate about the possible end of the era of GnRH-agonists, from what is known today, the advantages of GnRH-antagonists are most evident in our opinion.

A brief review of 2 lines of research and development of potent analogs of gonadotropin releasing hormone (GnRH) cyclic analogs and linear analogs intended to be safe long acting and hydrophilic is provided. Cyclic analogs of the linear peptide are highly conformationally constrained and hopefully resistant to enzymatic destruction therefore ...

GnRH antagonists in normal- responder patients
Fertility and Sterility, 2003To review the use of GnRH antagonists in normal-responding patients who are undergoing infertility treatment. Review article and case studies. For the normal-responding patient, GnRH antagonist protocols provide equivalent outcomes as GnRH agonist protocols, with the added patient benefit of significantly fewer treatment/injection days.

New GnRH antagonists are available in clinical practice. The different studies have confirmed the efficacy of these antagonists in preventing the LH surge. Two protocols have been described: in the multiple dose regimens, small doses of antagonist (0.25 mg) are injected starting on stimulation day 5 or 6 until hCG.

GnRH is a decapeptide, secreted from the hypothalamus in a pulsatile fashion, that regulates the synthesis and secretion of pituitary gonadotropins (1–9). In contrast, synthetic GnRH agonists or antagonists inhibit gonadotropin secretion and subsequently gonadal function (10, 11).

The pharmacology and the potential applications of GnRH antagonists
Current Opinion in Obstetrics and Gynecology, 1994Gonadotropin-releasing hormone (GnRH) antagonists are potent analogs of native GnRH. As biochemical probes, these recently developed compounds have allowed for profound insights into the physiology of the pituitary-ovarian and pituitary-testicular axes; as therapeutic alternatives, the GnRH antagonists hold great promise for various clinical ...
