Azacytidine prevents experimental xenogeneic graft-versus-host disease without abrogating graft-versus-leukemia effects

open access: yesOncoImmunology, 2017
The demethylating agent 5-azacytidine (AZA) has proven its efficacy in the treatment of myelodysplastic syndrome and acute myeloid leukemia. In addition, AZA can demethylate FOXP3 intron 1 (FOXP3i1) leading to the generation of regulatory T cells (Treg).

A Circuit of Mechanically Regulated Transcription Factors Balances Regenerative and Fibrotic Memory of Mesenchymal Stromal Cells

open access: yesAdvanced Science, EarlyView.
Producing MSCs on rigid culture substrates induces a scar‐making phenotype, jeapordizing therapeutic success. ‘Tissue‐soft’ surfaces prevent MSC fibrogenesis and preserve regenerative traits. An epigenetic network, driven by HOXA11 and SALL1, maintains ‘soft memory’ by keeping chromatin open in relaxed MSCs, promoting anti‐fibrotic programs.

Transfusion-associated graft-vs-host disease – A case report

open access: yes, 2016
Transfusion-associated graft-vs-host disease (TAGVHD) is a rare complication of blood transfusion. Unlike GVHD associated with hematopoietic stem cell transplantation TA-GVHD involves the patient's bone marrow and leads to bone marrow aplasia.

The IL-12 Cytokine and Receptor Family in Graft-vs.-Host Disease

open access: yesFrontiers in Immunology, 2019
Allogeneic hematopoietic cell transplantation (allo-HCT) is performed with curative intent for high- risk blood cancers and bone marrow failure syndromes; yet the development of acute and chronic graft-vs.-host disease (GVHD) remain preeminent causes of ...

Fully Humanized Bispecific T Cell Engager Shows Potent Activity in Central Nervous System and Peripheral Tumors

open access: yesAdvanced Science, EarlyView.
This study reports the development of a fully humanized bispecific T cell engager targeting IL13RA2, a tumor‐associated antigen enriched in glioblastoma. This off‐the‐shelf immunotherapy drives potent, antigen‐dependent T cell activation and tumor killing, and prolongs survival in experimental GBM and other solid tumors models without detectable off ...

Ocular Graft versus Host Disease

open access: yes, 2015
Allogeneic hematopoietic stem cell transplantation is considered the standard of care for hematological and lymphoid malignancies. One of the complications associated to this therapy is an immune-mediated reaction known as graft versus host disease with ...

Cytokine Detection and Modulation in Acute Graft vs. Host Disease in Mice

open access: yesMediators of Inflammation, 1994
A murine model for acute lethal graft vs. host disease (GVHD) was used to study the role that a number of cytokines play in the development of lethal GVHD. In this study we focused on the role of IL-1, IL-2, IL-4, IL-6, IFN-γ and TNF-α.

Treatment of Severe Steroid-Refractory Acute-Graft-vs.-Host Disease With Mesenchymal Stem Cells–Single Center Experience

open access: yesFrontiers in Bioengineering and Biotechnology, 2018
The most effective treatment of steroid refractory acute graft vs. host disease (aGvHD) is not yet established and mesenchymal stem cells (MSC) appear to be a promising therapy for the condition. We report single center case series of three patients, who

Engineering Approaches to Modify Immunomodulatory Functions of Mesenchymal Stromal Cells (MSCs): Tissue Regeneration and Clinical Application

open access: yesAdvanced Science, EarlyView.
Mesenchymal stromal cells (MSCs) show promise for treating immune‐related disorders through immunomodulation and tissue regeneration. This review gives a brief overview of current clinical approval of MSC therapies. It also discussed how bioengineering, including genetic modification, biomaterial delivery, extracellular vesicles, and iPSC‐derived MSCs,
