Monoclonal antibodies for detection, serological characterization and immunopurification of grapevine fleck virus

Ten stable hybridoma cell lines secreting monoclonal antibodies to grapevine fleck virus (GFkV) were selected after fusing spleen cells of immunized Balb/C mice with mouse myeloma cells (SP2/0-Ag 14). All MAbs reacted positively in ELISA with leaf extracts from fifty GFkV-infected grapevines from various geographical origins.

AbstractAlthough Grapevine fleck virus (GFkV) has a worldwide distribution, data about its molecular variability are very scarce. A genome region encoding the central part of the capsid protein gene was amplified from 36 GFkV isolates recovered from a relatively restricted area (Slovakia and the Czech Republic).

SummaryA region encoding the putative RNA replicase domain of Grapevine fleck virus (GFkV) was amplified from infected grapevine samples using the reverse transcription‐polymerase chain reaction (RT‐PCR). Two different sizes of virus specific PCR products were consistently detected in different grapevine varieties using the same pair of primers.
