Inference in group sequential designs with causal mechanisms: implications for power and mediation analysis

open access: yesBMC Medical Research Methodology
Background Group sequential designs are increasingly employed to allow trials to stop early with statistical rigor. While existing work focuses on intention-to-treat effect on clinical endpoints, the properties of mediation analysis (commonly conducted ...

Investigating transcription factor dynamics in health and disease using FRAP

open access: yesFEBS Letters, EarlyView.
FRAP analysis of GFP‐tagged transcription factors reveals how molecular mobility and target engagement change in response to drug treatment. By combining live‐cell imaging, quantitative model fitting, and statistical analysis, this approach uncovers transcription factor dynamics linked to disease mechanisms, providing a powerful framework for ...

Bayesian evaluation of group sequential clinical trial designs

open access: yesStatistics in Medicine, 2006
AbstractClinical trial designs often incorporate a sequential stopping rule to serve as a guide in the early termination of a study. When choosing a particular stopping rule, it is most common to examine frequentist operating characteristics such as type I error, statistical power, and precision of confidence intervals (Statist. Med. 2005, in revision).

Conserved binding mode but diverse interfaces of MreC‐PBP2 interactions

open access: yesFEBS Letters, EarlyView.
The crystal structure of abMreC reveals a conserved two β‐barrel architecture and provides structural insights into its role within the bacterial elongasome. The abMreC–abPBP2 complex model identifies the molecular basis of MreC‐mediated PBP2 recognition, contributing to the regulation of peptidoglycan synthesis.

Microbiome‐blood–brain barrier interactions in aging — mechanisms and therapeutic potential

open access: yesFEBS Letters, EarlyView.
Aging reshapes the gut microbiome (↓SCFA‐producing commensals; ↑pro‐inflammatory outputs), shifting circulating metabolites (↓SCFAs; ↑LPS, ↑TMAO, ↑PAA) that act at the BBB to increase nonspecific transcytosis, alter transport, and promote astrocyte reactivity, heightening brain vulnerability.

Practical and analytical considerations when performing interim analyses in diagnostic test accuracy studies

open access: yesDiagnostic and Prognostic Research
Interim analysis is a common methodology in randomised clinical trials but has received less attention in studies of diagnostic test accuracy. In such studies, early termination for futility may be beneficial if early evidence indicates that a diagnostic

Drilling down to success: Comparative insight on single-drill and sequential-drill techniques for dental implant osseointegration using osteoprotegerin levels - A randomized control trial

open access: yesJournal of Oral Biology and Craniofacial Research
Background: To evaluate and compare osteoblastic activity and osseointegration between single-drill and sequential-drill implant site preparation techniques, using osteoprotegerin (OPG) levels as a biomarker. Methods: A randomized controlled trial with a
