Distinct CTC Specific RNA Profile Enables NSCLC Early Detection and Dynamic Monitoring of Advanced NSCLC

open access: yesAdvanced Science, EarlyView.
A quantitative CTC RNA assay is developed by incorporating multi‐antibody‐based CTC isolation and specific mRNA quantification using RT‐ddPCR. The NSCLC CTC ScoreD demonstrates high accuracy for early‐stage NSCLC detection, significantly outperforming serum CEA. NSCLC CTC ScoreM exhibits a more accurate early‐warning of responses to different therapies

PABPN1 Couples the Polyadenylation and Translation of Maternal Transcripts to Mouse Oocyte Meiotic Maturation

open access: yesAdvanced Science, EarlyView.
This study reveals that cytoplasmic PABPN1 is essential for mouse oocyte meiotic maturation by coordinating polyadenylation, translation, and degradation of maternal mRNAs. Pabpn1 knockout disrupts CDK1 activation, spindle formation, and chromosome alignment by impairing maturation‐promoting factor (MPF) regulation and BTG4‐mediated deadenylation ...

SMC4 Promotes Prostate Cancer Cell Proliferation and Metastasis via the Rheb/mTOR Pathway

open access: yesAdvanced Science, EarlyView.
Structural maintenance of chromosome protein 4 (SMC4) has been implicated in prostate cancer metastasis. Knockdown of SMC4 in RM1‐LM cells significantly attenuates proliferation, migration, and lung metastasis in vivo. Furthermore, SMC4 interacts with GLUT1 (Slc2a1), thereby enhancing glycolysis, ATP production, and activation of the Rheb/mTOR pathway.

Light‐Triggered CRISPR/Cas12a for Genomic Editing and Tumor Regression

open access: yesAngewandte Chemie, EarlyView.
A photo‐triggered CRISPR/Cas12a machinery for gene editing is introduced. The machinery is applied for endogenous DNA methyltransferase 1 (DNMT1) gene editing and for in vitro disruption of the hepatocyte growth factor (HGF) gene. The machinery is used for in vivo knockout and disruption of the HGF gene in HepG2 tumors, resulting in effective apoptosis

The INAVA mRNA in Extracellular Vesicles Activates Normal Ovarian Fibroblasts by Phosphorylation–Ubiquitylation Crosstalk of HMGA2

open access: yesAdvanced Science, EarlyView.
Ovarian cancer‐derived extracellular vesicles (EVs) transfer INAVA mRNA to normal ovarian fibroblasts (NOFs). Translated INAVA protein inhibits HMGA2 phosphorylation‐dependent ubiquitylation and proteasomal degradation, promoting NOF activation.

Placental RTN3L‐dependent ER‐Phagy Contributes to Fetal Testicular Dysplasia Upon Environmental Stress

open access: yesAdvanced Science, EarlyView.
Gestational environmental stress activates ER‐phagy by increasing placental Rtn3l mRNA N6‐methyladenosine modification, further inhibiting placental estradiol synthesis and contributing to fetal testicular dysplasia. This study demonstrates the early prevention and treatment of adult male infertility from the perspective of fetal‐derived diseases ...

FABP4 as a Mediator of Lipid Metabolism and Pregnant Uterine Dysfunction in Obesity

open access: yesAdvanced Science, EarlyView.
Obesity during late pregnancy contributes to uterine smooth muscle dysfunction, but the underlying mechanisms are unclear. This study identifies fatty acid binding protein 4 (FABP4) as a key player in the process, mediating excessive fatty acid uptake, lipid accumulation, and mitochondrial dysfunction in myometrial cells. FABP4 could be a novel uterine
