PepTune: De Novo Generation of Therapeutic Peptides with Multi-Objective-Guided Discrete Diffusion [PDF]

We present PepTune, a multi-objective discrete diffusion model for simultaneous generation and optimization of therapeutic peptide SMILES. Built on the Masked Discrete Language Model (MDLM) framework, PepTune ensures valid peptide structures with a novel bond-dependent masking schedule and invalid loss function.

This study introduces a novel therapeutic approach that employs a conductive hydrogel (HEPP) responsive to near‐infrared light, pH levels, and reactive oxygen species. Its goal is to enhance the healing environment affected by elevated glucose levels, insufficient oxygen, and bacterial infections in diabetes‐related wounds.

Polymer Brush‐Based Asymmetric Porous Patch with Excellent Anti‐Bacterial and Anti‐Contamination Properties Enables Infectious Abdominal Wall Defect Repair

A zwitterionic asymmetric patch is developed via self‐induced phase separation. The cationic layer exhibits anti‐bacterial efficacy and releases pro‐healing factor, while the zwitterionic hydrogel prevents contamination and visceral adhesion. The strong mechanical interlocking of two layers leads to a high and stable burst pressure tolerance.
