Reclassification of BRCA2 Variants of Uncertain Significance Using Saturation Genome Editing Combined with Clinical Phenotypes in Breast Cancer

open access: yesCurrent Oncology
Purpose: To evaluate the pathogenicity of BRCA2 variants of uncertain significance (VUS) located within the functionally critical exons 15–26 using published SGE data, and to reclassify these VUS by integrating clinical phenotypes. Methods: A total of 15,

Peripheral lysosomes recruit PLEKHG3 to focal adhesions and restrain protrusion dynamics

open access: yesFEBS Letters, EarlyView.
Proximity‐dependent labeling at the LAMTOR complex revealed the Rho GEF PLEKHG3 as a lysosome‐proximal protein directing the study toward the influence of lysosome positioning on actin dynamics and cell motility. We show that PLEKHG3 colocalizes with lysosomes at focal adhesion sites and observe that forced peripheral dispersion of lysosomes hinders ...

Cancer Trends: Cancer of unknown primary

open access: yes, 2019
Between 2012 and 2016, over 450 patients per year were diagnosed with a malignant cancer where the primary site of their cancer was unspecified. These cancers were identified by the presence of metastatic disease where the cancer has spread to other ...

Hereditary Cancer Model of Care: HSE National Cancer Control Programme

open access: yes, 2023
Genetics is now recognised as a growing and integral part of cancer care, with a role across treatment, early detection and risk reduction. Identification of cancer predisposition will only lead to patient benefit, if necessary services are in place and ...

Fruits, vegetables, and hMLH1 protein-deficient and -proficient colon cancer: The Netherlands cohort study. [PDF]

open access: yes, 2005
BACKGROUND: Clinical and pathologic differences exist between colon carcinomas deficient and proficient in the mismatch repair protein hMLH1. Animal and in vitro studies suggest that fruits, vegetables, folate, and antioxidants are associated with ...

Cancer Trends: Cancer of the liver and biliary tract

open access: yes, 2010
Liver cancer consists of two main types—hepatocellular carcinoma (HCC) which accounts for about 60% of cases, and intrahepatic bile duct cholangiocarcinoma (ICC), which accounts for about 20%. The liver is one of the less common cancer sites, about 0.

A context‐dependent modulatory role for eIF6 in acquired resistance to vemurafenib in melanoma

open access: yesFEBS Letters, EarlyView.
Acquired resistance to vemurafenib upregulates the translation factor eIF6 in melanoma cells. Silencing eIF6 in resistant cells reduces proliferation and partially restores drug sensitivity, whereas its overexpression increases sensitivity across melanoma lines regardless of BRAF status, via modulation of mTOR, S6K, and MAPK signaling.
