IKKβ and USP28 Regulate HEY1 Stability to Promote Cancer Stemness and Immune Evasion in Hepatocellular Carcinoma

open access: yesAdvanced Science, EarlyView.
The uncovered IKKβ‐USP28‐HEY1 axis fuels cancer stemness and immune evasion in hepatocellular carcinoma. USP28 deubiquitinates HEY1 upon IKKβ‐mediated phosphorylation, conferring PD‐1/PD‐L1 blockade resistance. Pharmacological inhibition of USP28 sensitizes resistant tumors to anti‐PD‐1 immunotherapy, revealing a promising therapeutic strategy ...

Artificial Intelligence‐Based Body Composition Analysis Reveals Sex‐Specific Prognostic Markers and Their Clinical Value in Gastric Cancer: A Multicenter Study

open access: yesAdvanced Science, EarlyView.
This study applied AI to quantify multidimensional body composition from CT images in gastric cancer and healthy controls. Distinct sex‐specific patterns and disease‐related alterations were identified and were associated with survival. Higher muscle and fat measures were linked to improved outcomes.

Discovery and Engineering of a Rat Endogenous Retrovirus Reverse Transcriptase for Efficient Prime Editing

open access: yesAdvanced Science, EarlyView.
We screened 558 reverse transcriptases and engineered an optimized rat endogenous retrovirus‐derived variant, enRERV‐RT, via structure‐guided engineering and deep mutational scanning. This enhanced prime editor, based on the engineered RT, outperforms conventional M‐MLV‐RT systems across plant and animal cells, particularly at hard‐to‐edit loci ...

Answers to modal and hypothetical questions in knowledge base systems

Applied Intelligence, 1992
This paper describes a computational model which is geared towards providing helpful answers to modal and hypothetical questions in knowledge base systems (KBSs). The model is an essential component of a more complete model which aims at giving helpful answers to questions presented in a natural language.
