A Deep Representation Learning Method for Quantitative Immune Defense Function Evaluation and Its Clinical Applications

ImmuDef, a novel algorithm to quantitatively evaluate the anti‐infection immune defense function of an individual based on RNA‐seq data via a variational autoencoder (VAE) model. It is validated on 3200+ samples across four immune states with high accuracy. It can serve as a metric for disease severity and prognosis across pathogenic cohorts.

SLC2A3‐Mediated Lactate Metabolism Promotes Lung Cancer Bone Metastasis by Modulating P53 Lactylation and Immune Evasion

SLC2A3 derived lactate promotes metastasis through p53 lactylation at K120 and osteoclast differentiation. Pharmacological inhibition of SLC2A3 upregulates PD‐1 expression on CD8+ T cells via lactate induced p53 lactylation, which modulates immune evasion. Following intracerebral inoculation of adult Balb/c Byj mice, the MHV-4 strain of mouse hepatitis virus (MHV) had an LD50 of less than 0.1 PFU, whereas its monoclonal antibody resistant variant V5A13.1 had an LD50 of 10(4.2) PFU.

NK Cell Activation by Platinum Boosts Immunotherapy in HR+/HER2− Breast Cancer

Multi‐omics and single‐cell analyses identify activated NK cells as key mediators of anti‐PD‐(L)1 response in HR+/HER2− breast cancer. Platinum chemotherapy enhances NK cell cytotoxicity through NF‐κB–associated signaling, synergizing with immunotherapy and providing a rationale for combination treatment.
