Synthetic Lethality of SHP2 and XIAP Suppresses Proliferation and Metastasis in KRAS‐mutant Nonsmall Cell Lung Cancer

open access: yesAdvanced Science, Volume 12, Issue 15, April 17, 2025.
Embelin cotargets SHP2 to disrupt the SHP2/GRB2/SOS complex, inhibiting cancer‐related signaling pathways such as MAPK, (PI3K)/AKT pathway, and XIAP to induce apoptosis signaling pathway, respectively, leading to synthetic lethality in KRAS‐mutant NSCLC cells both in vivo and in vitro.

HUWE1‐Mediated Degradation of MUTYH Facilitates DNA Damage and Mitochondrial Dysfunction to Promote Acute Kidney Injury

open access: yesAdvanced Science, Volume 12, Issue 13, April 3, 2025.
MutY homolog (MUTYH) protein levels are significantly decreased in the kidneys of acute kidney injury (AKI) patients. The upregulation of the E3 ligase HUWE1 mediates the degradation of MUTYH in renal tubular epithelial cells, exacerbating DNA damage in both the nucleus and mitochondria, thereby promoting AKI. Targeting the HUWE1/MUTYH pathway may be a

AIRR-C IG Reference Sets: curated sets of immunoglobulin heavy and light chain germline genes

open access: yesFrontiers in Immunology
IntroductionAnalysis of an individual’s immunoglobulin (IG) gene repertoire requires the use of high-quality germline gene reference sets. When sets only contain alleles supported by strong evidence, AIRR sequencing (AIRR-seq) data analysis is more ...

DNA‐PKcs‐Driven YAP1 Phosphorylation and Nuclear Translocation: a Key Regulator of Ferroptosis in Hyperglycemia‐Induced Cardiac Dysfunction in Type 1 Diabetes

open access: yesAdvanced Science, EarlyView.
In the context of chronic hyperglycemia, a DDR is initiated, leading to the pathological activation of DNA‐PKcs in the diabetic heart. This activated DNA‐PKcs directly interacts with and phosphorylates YAP1 at Thr226, thereby increasing the nuclear expression of YAP1.

NF-KB protein purification from bovine spleen: Nucleotide stimulation and binding site specificity [PDF]

open access: yes, 1988
The activity of the enhancer for the κ immunoglobulin light chain gene critically depends on the presence in the nucleus of the NF-κB protein. We purified NF-κB over 50,000-fold and identified two protein species, 42 and 44 kDa, that could be eluted and ...
