Senescent Synovial Intimal Fibroblasts Aggravate Osteoarthritis by Regulating Macrophage Polarization and Chondrocyte Phenotype Through ANGPTL4‐α5β1 Axis

open access: yesAdvanced Science, EarlyView.
Senescent synovial intimal fibroblasts (SIF) are identified as key drivers of osteoarthritis. They promote M1 macrophage polarization and cartilage degeneration via the ANGPTL4–α5β1 axis, regulated by transcription factors EGR1 and ATF3. Pharmacological inhibition of this pathway alleviates disease, revealing SIF senescence as a promising therapeutic ...

Prediction of outcomes after cardiac arrest by a generative artificial intelligence model

open access: yesResuscitation Plus
Aims: To investigate the prognostic accuracy of a non-medical generative artificial intelligence model (Chat Generative Pre-Trained Transformer 4 - ChatGPT-4) as a novel aspect in predicting death and poor neurological outcome at hospital discharge based

Single‐Cell Mitochondrial Lineage Tracing Decodes Fate Decision and Spatial Clonal Architecture in Human Hematopoietic Organoids

open access: yesAdvanced Science, EarlyView.
This study repurposes mitochondrial DNA mutations as endogenous barcodes for lineage tracing in human pluripotent stem cell‐derived organoids. Integrated with transcriptomic and spatial data, it reveals NOTCH‐mediated stromal‐progenitor crosstalk orchestrates clonal dynamics and spatial zonation during early hematopoietic development, offering a non ...

A Programmable Nanoreactor Orchestrates Cascade of DNA Sensing to Amplify cGAS‐STING Activation for Cancer Immunotherapy

open access: yesAdvanced Science, EarlyView.
A programmable nanoreactor was engineered for precise cGAS‐STING pathway activation in cancer immunotherapy. This strategy bridges targeted tumor DNA damage with DC immunomodulation, providing a translatable platform to unlock innate and adaptive antitumor immunity.

SIRT5–RAC2 Axis Drives Monocyte‐to‐Macrophage Differentiation to Promote Inflammatory Injury in Premature Ovarian Insufficiency

open access: yesAdvanced Science, EarlyView.
SIRT5 desuccinylates and stabilizes RAC2, activating CSF1R‐dependent signaling to drive monocyte differentiation into M0 macrophages and their polarization toward pro‐inflammatory M1 phenotypes in CTX‐induced premature ovarian insufficiency. Inhibiting the SIRT5‐RAC2 axis attenuates inflammation, reduces granulosa cell apoptosis, and preserves ...

Microgel‐Based Hierarchical Porous Hydrogel Patch with Adhesion and Resilience for Myocardial Infarction

open access: yesAdvanced Science, EarlyView.
This study develops a hierarchically porous hydrogel patch strategy (HPMP), based on gas‐shearing microfluidics and an aqueous two‐phase system to fabricate porous microgels as microgel‐based bioinks. The porous microgels with controllable porous structure exhibit excellent cellular behavior.

Post cardiac arrest left ventricular ejection fraction associated with survival to discharge

open access: yesResuscitation Plus
Background: Post cardiac arrest left ventricular ejection fraction (LVEF) is routinely assessed, but the implications of this are unknown. This study aimed to assess the association between post cardiac arrest LVEF and survival to hospital discharge ...
