Pharmacokinetics of Drugs That Inactivate Metabolic Enzymes

Journal of Pharmaceutical Sciences, 1991
The pharmacokinetics of drugs that cause inactivation of metabolic enzymes were derived for the single-compartment model with linear elimination kinetics. A simple mechanism for suicide inactivation of enzyme by its substrate was used to model the relationship between elimination of drug and enzyme activity as a function of time.

Metabolic inactivation of mutagens in Drosphila melanogaster

Mutation Research - Fundamental and Molecular Mechanisms of Mutagenesis, 1988
The route of administration of a drug is a pharmacological factor to be reckoned with. In Drosophila, a whole-animal object for mutagenicity studies, the way in which a mutagen is applied can also be of crucial importance. In this study the mutagenicity of a number of directly acting agents was determined after feeding or injection of the mutagen ...

Formate metabolism in the cobalamin‐inactivated rat

British Journal of Haematology, 1990
SummaryEndogenous formate levels in blood and liver were assayed in rats both after inactivation of cobalamin (Cbl) by exposure to N2O as well as in air‐breathing controls. The uptake of [14C]formate by tetrahydrofolate (H4folate) in bone marrow cells and liver homogenate and the incorporation of [14C]formate into purine, pyrimidine, methionine, serine

Piezo-catalytic persulfate activation system for water advanced disinfection: Process efficiency and inactivation mechanisms

, 2020
This study developed a novel method based on piezoelectric catalytic persulfate (PS) activation for water advanced disinfection. Silver modified barium titanate (Ag-BTO) was synthesized and utilized as a piezoelectric catalyst to activate PS under ...

Metabolic activation and inactivation of chemical carcinogens

Journal of Toxicology and Environmental Health, 1980
Chemical carcinogens are metabolized by numerous pathways catalyzed by enzymes in endoplasmic reticulum and other parts of the cell. Reactions in which functional groups are created (epoxidation and epoxide hydration, catalyzed by cytochrome P-450-linked monooxygenase and epoxide hydratase, respectively) are especially important in the activation of ...

Inactivation of CYP2A6 and CYP2A13 during Nicotine Metabolism

The Journal of Pharmacology and Experimental Therapeutics, 2006
Nicotine is the major addictive agent in tobacco. The primary catalyst of nicotine metabolism in humans is CYP2A6. However, the closely related enzyme CYP2A13 is a somewhat better catalyst. CYP2A13 is an extrahepatic enzyme that is an excellent catalyst of the metabolic activation of the tobacco-specific carcinogen 4-(methylnitrosamine)-1-(3-pyridyl)-1-

Metabolic inactivation of bleomycin analogs by bleomycin hydrolase

Pharmacology & Therapeutics, 1988

Tyrosine transaminase: Inactivation by tyrosine metabolic end products

Biochimica et Biophysica Acta (BBA) - Enzymology, 1970
Abstract The tyrosine metabolites, p- hydroxyphenylpyruvate , homogentisate, and fumarylacetoacetate were examined as inhibitors and inactivators of purified rat liver tyrosine α-ketoglutarate transaminase. Of these, homogentisate proved to be a very powerful inactivator when the enzyme was preincubated with it. This effect was blocked by prior

inactivation in vitro of microsomal oxidases during parathion metabolism

Biochemical Pharmacology, 1978
Abstract The inactivation in vitro of microsomal oxidases during parathion metabolism was examined in relation to the binding to the microsomal membrane of sulfur resulting from desulfuration. Modulation of sulfur binding with sulfhydryl compounds revealed that the majority of bound sulfur is unrelated to inactivation.
