New Functions of the Inositol Polyphosphate 5-Phosphatases in Cancer
Current Pharmaceutical Design, 2016Inositol polyphosphate 5-phosphatases act on inositol phosphates and phosphoinositides as substrates. They are 10 different isoenzymes and several splice variants in the human genome that are involved in a series of human pathologies such as the Lowe syndrome, the Joubert and MORM syndromes, breast cancer, glioblastoma, gastric cancer and several other

Regulation of second messengers by the inositol polyphosphate 5-phosphatases
Biochemical Society Transactions, 1996The inositol polyphosphate 5-phosphatases are an enlarging family of enzymes that terminate the signals generated by the phosphoinositide kinases and phospholipase C. Given the diverse signalling functions of both the polyphosphoinositides and Ins(1,4,5)P3, it is predicted that the 5-phosphatases will play a critical role in regulating many cellular ...

Mutation of the Conserved Domains of Two Inositol Polyphosphate 5-Phosphatases
Biochemistry, 1996Two short amino acid motifs, WXGDXNXR and PXWCDRXL, define a large family of inositol polyphosphate 5-phosphatases. We tested the importance of seven of these conserved amino acids to substrate binding and catalysis by mutating each to alanine in the platelet 75 kDa inositol polyphosphate 5-phosphatase II (5-phosphatase II).

Pharbin, a Novel Inositol Polyphosphate 5-Phosphatase, Induces Dendritic Appearances in Fibroblasts
Biochemical and Biophysical Research Communications, 1999We have cloned a cDNA encoding a novel protein pharbin with a homology to inositol polyphosphate 5-phosphatases. Pharbin contains relatively well-conserved catalytic motifs for 5-phosphatase, a proline-rich sequence corresponding to the SH3-binding motif, and a sequence consistent with the CaaX motif at the C-terminus.

Activation of the 43 kDa Inositol Polyphosphate 5-Phosphatase by 14-3-3ΞΆ
Biochemistry, 1997The 43 kDa inositol polyphosphate 5-phosphatase (5-phosphatase) hydrolyzes and thereby inactivates the second messenger molecules inositol 1,4,5-trisphosphate -Ins(1,4,5)P3- and inositol 1,3,4,5-tetrakisphosphate in a signal terminating reaction.

The phosphatidylinositol phosphate signaling pathway is involved in many crucial cellular functions. The myo-inositol polyphosphate 5-phosphatases (5PTases) (E.C. 3.1.3.56) comprise a large protein family that hydrolyze 5-phosphates from a variety of phosphatidylinositol phosphate and inositol phosphate substrates.

The prognostic value of inositol polyphosphate 5-phosphatase in cutaneous squamous cell carcinoma
Journal of the American Academy of Dermatology, 2019Inositol polyphosphate 5-phosphatase (INPP5A) has been shown to play a role in development and progression of cutaneous squamous cell carcinoma (cSCC). The goal of the current study was to explore the prognostic value of INPP5A expression in cSCC.A total of 189 cases of actinic keratosis and SCC in 174 patients were identified; clinical and outcome ...

Regulation of PtdIns(3,4,5)P3/Akt signalling by inositol polyphosphate 5-phosphatases
Biochemical Society Transactions, 2016The phosphoinositide 3-kinase (PI3K) generated lipid signals, PtdIns(3,4,5)P3 and PtdIns(3,4)P2, are both required for the maximal activation of the serine/threonine kinase proto-oncogene Akt. The inositol polyphosphate 5-phosphatases (5-phosphatases) hydrolyse the 5-position phosphate from the inositol head group of PtdIns(3,4,5)P3 to yield PtdIns(3,4)

The maize brevis plant1 is a type II inositol polyphosphate 5-phosphatase
Journal of Plant Biochemistry and Biotechnology, 2017The genes associated with the dwarf phenotype have been utilized in crop breeding to prevent lodging and stem breakage. Brevis plant1 (Bv1), encoding a putative inositol polyphosphate 5-phosphatase (5PTase), has been associated with stem elongation in maize (Zea mays L.); however, the enzymatic activity of BV1 has not been experimentally characterized.
