Peritoneal spread of tumors is a major problem in cancer management. Patients develop a marked deterioration in quality of life and shortened survival. This is in part due to bowel obstructions, marked ascites, and overall increase debilitation. Standard medical management has shown to be inadequate for the treatment of these problems.

By engulfing apoptotic cancer cells, macrophages upregulate ODC1 and produce putrescine, which elevates osteopontin (OPN) expression. OncoLRC, a one‐component lipid‐mRNA complex, enables efficient spleen‐targeted delivery at an exceptionally low lipid‐to‐mRNA mass ratio (1.5:1), robustly activates immune responses, inhibits tumor growth, and synergizes with checkpoint blockade, presenting a next‐generation platform for mRNA vaccines.
