The neural crest‐associated gene ERRFI1 is involved in melanoma progression and resistance toward targeted therapy

open access: yesMolecular Oncology, EarlyView.
ERRFI1, a neural crest (NC)‐associated gene, was upregulated in melanoma and negatively correlated with the expression of melanocytic differentiation markers and the susceptibility of melanoma cells toward BRAF inhibitors (BRAFi). Knocking down ERRFI1 significantly increased the sensitivity of melanoma cells to BRAFi.

Bias Reduction in Robust Mean–Geometric Mean Linking via SIMEX

open access: yesAlgorithms
Robust mean–geometric mean (MGM) linking is a method for comparing the performance of two groups on a test involving dichotomous items and is particularly suited to settings with fixed and sparse differential item functioning (DIF).

In vitro models of cancer‐associated fibroblast heterogeneity uncover subtype‐specific effects of CRISPR perturbations

open access: yesMolecular Oncology, EarlyView.
Development of therapies targeting cancer‐associated fibroblasts (CAFs) necessitates preclinical model systems that faithfully represent CAF–tumor biology. We established an in vitro coculture system of patient‐derived pancreatic CAFs and tumor cell lines and demonstrated its recapitulation of primary CAF–tumor biology with single‐cell transcriptomics ...

Extensions to Mean–Geometric Mean Linking

open access: yesMathematics
Mean-geometric mean (MGM) linking is a widely used method for linking two groups within the two-parameter logistic (2PL) item response model. However, the presence of differential item functioning (DIF) can lead to biased parameter estimates using the ...

The Estimation of Item Response Models with the lmer Function from the lme4 Package in R [PDF]

open access: yes
In this paper we elaborate on the potential of the lmer function from the lme4 package in R for item response (IRT) modeling. In line with the package, an IRT framework is described based on generalized linear mixed modeling. The aspects of the framework

Dual targeting of RET and SRC synergizes in RET fusion‐positive cancer cells

open access: yesMolecular Oncology, EarlyView.
Despite the strong activity of selective RET tyrosine kinase inhibitors (TKIs), resistance of RET fusion‐positive (RET+) lung cancer and thyroid cancer frequently occurs and is mainly driven by RET‐independent bypass mechanisms. Son et al. show that SRC TKIs significantly inhibit PAK and AKT survival signaling and enhance the efficacy of RET TKIs in ...

Estimation of Standard Error, Linking Error, and Total Error for Robust and Nonrobust Linking Methods in the Two-Parameter Logistic Model

open access: yesStats
The two-parameter logistic (2PL) item response theory model is a statistical model for analyzing multivariate binary data. In this article, two groups are brought onto a common metric using the 2PL model using linking methods. The linking methods of mean–

Detection of circulating tumor DNA in colorectal cancer patients using a methylation‐specific droplet digital PCR multiplex

open access: yesMolecular Oncology, EarlyView.
We developed a cost‐effective methylation‐specific droplet digital PCR multiplex assay containing tissue‐conserved and tumor‐specific methylation markers. The assay can detect circulating tumor DNA with high accuracy in patients with localized and metastatic colorectal cancer.

Bias-Corrected Fixed Item Parameter Calibration, with an Application to PISA Data

open access: yesStats
Fixed item parameter calibration (FIPC) is commonly used to compare groups or countries using an item response theory model with a common set of fixed item parameters.
