Lilrb4a Suppression Reprograms Microglia to Mitigate APOE4‐Associated Amyloid Plaques and Cerebral Amyloid Angiopathy in Association With a PPAR‐Linked Pro‐Clearance State

open access: yesAdvanced Science, EarlyView.
Targeting Lilrb4a in Apolipoprotein E4 (APOE4)‐associated Alzheimer's disease (AD) reprograms microglia toward a beneficial, phagocytic state. Genetic deletion or antisense inhibition of Lilrb4a suppresses p‐SHP2/NF‐κB/STAT1 signaling, restores PPAR‐linked lipid and energy metabolism, and reduces amyloid plaque burden and cerebral amyloid angiopathy ...

Rapid resolution of non-segmental vitiligo in a patient treated with abrocitinib: A case report

open access: yesSAGE Open Medical Case Reports
Vitiligo is a common, autoimmune, depigmenting disorder of the skin. Janus Kinase inhibitors have emerged as promising topical and oral therapeutic options for vitiligo.

c-Src binds to the cancer drug Ruxolitinib with an active conformation. [PDF]

open access: yesPLoS ONE, 2014
The cancer drug Ruxolitinib is a potent janus kinase inhibitor approved for the treatment of the myeloproliferative neoplasms. In addition, Ruxolitinib has weak inhibitory activity against a panel of other kinases, including Src kinase.

The rationale for Janus kinase inhibitors for the treatment of spondyloarthritis [PDF]

open access: yesRheumatology, 2018
The pathogenesis of SpA is multifactorial and involves a range of immune cell types and cytokines, many of which utilize Janus kinase (JAK) pathways for signaling. In this review, we summarize the animal and pre-clinical data that have demonstrated the effects of JAK blockade on the underlying molecular mechanisms of SpA and provide a rationale for JAK

Triple‐Negative Breast Cancer Cells Utilize IL8 and CXCL1 to Suppress NK Cells’ Function and Facilitate Cancer Metastasis

open access: yesAdvanced Science, EarlyView.
Triple‐negative breast cancer (TNBC) cells evade natural killer (NK) cell immunity by secreting IL8 and CXCL1. These chemokines suppress NK cells’ function via CXCR1/2 and enhance cancer cells’ survival through PD‐L1 upregulation and BCL‐2 anti‐apoptotic signaling.

Indirect inhibition of the NLRP3-interleukin-1β axis contributes to the efficacy of JAK1 inhibitors in experimental colitis and human ulcerative colitis

open access: yesNature Communications
Tofacitinib, a pan–Janus kinase inhibitor, and the Janus kinase 1–preferential inhibitors Upadacitinib and Filgotinib are approved for the treatment of ulcerative colitis, yet their molecular mechanisms of action remain incompletely understood.

Janus kinase inhibitors for the treatment of COVID-19

open access: yesCochrane Database of Systematic Reviews, 2022
With potential antiviral and anti-inflammatory properties, Janus kinase (JAK) inhibitors represent a potential treatment for symptomatic severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection. They may modulate the exuberant immune response to SARS-CoV-2 infection. Furthermore, a direct antiviral effect has been described.

RNA‐Binding Protein RBM25 Targets the mRNA Stability of GTPase Rab22a to Restrict Viral Entry and Infection

open access: yesAdvanced Science, EarlyView.
This study identifies RNA‐binding protein RBM25 as a broad‐spectrum antiviral factor acting independently of type I interferon. It blocks viral entry by suppressing GTPase Rab22a via the RC3H1‐mediated destabilization of Rab22a mRNA. Viral downregulation of RBM25 enhances GTPase Rab22a expression and viral entry, revealing an unreported post ...

Deregulated expression of Cdc6 as BCR/ABL-dependent survival factor in chronic myeloid leukemia cells

open access: yesTumor Biology, 2017
Chronic myeloid leukemia is characterized by the presence of the reciprocal translocation t(9;22) and the BCR/ABL oncogene. The BCR/ABL oncogene activates multiple signaling pathways and involves the dysregulation of oncogenes during the progression of ...

Upadacitinib-induced paradoxical face and scalp dermatitis: A case report of a novel sequela

open access: yesSAGE Open Medical Case Reports, 2023
Atopic dermatitis is a chronic, pruritic inflammatory cutaneous condition that can carry significant morbidity. Severe or recalcitrant atopic dermatitis is often treated with immunosuppressants, biologics, or immune-modulating small molecule therapies ...
