Confocal Raman data analysis enables identifying apoptosis of MCF-7 cells caused by anticancer drug paclitaxel [PDF]

open access: yes, 2013
Confocal Raman microscopy is a noninvasive, label-free imaging technique used to study apoptosis of live MCF-7 cells. The images are based on Raman spectra of cells components, and their apoptosis is monitored through diffusion of cytochrome c in ...

Label-free nonlinear optical imaging of mouse retina

open access: yesBiomedical Optics Express, 2015
A nonlinear optical (NLO) microscopy system integrating stimulated Raman scattering (SRS), two-photon excited fluorescence (TPEF) and second-harmonic generation (SHG) was developed to image fresh mouse retinas. The morphological and functional details of various retinal layers were revealed by the endogenous NLO signals.

β‐TrCP overexpression enhances cisplatin sensitivity by depleting BRCA1

open access: yesMolecular Oncology, EarlyView.
Low levels of β‐TrCP (Panel A) allow the accumulation of BRCA1 and CtIP, which facilitate the repair of cisplatin‐induced DNA damage via homologous recombination (HR) and promote tumor cell survival. In contrast, high β‐TrCP expression (Panel B) leads to BRCA1 and CtIP degradation, impairing HR repair, resulting in persistent DNA damage and apoptosis ...

Intravital imaging by simultaneous label-free autofluorescence-multiharmonic microscopy

open access: yesNature Communications, 2018
Label-free and real-time visualization of the tumor microenvironment is attractive but challenging. Here the authors present an approach for simultaneous autofluorescence functional imaging and second/third harmonic generation imaging of structural ...

Handheld optical-resolution photoacoustic microscopy [PDF]

open access: yes, 2017
Optical-resolution photoacoustic microscopy (OR-PAM) offers label-free in vivo imaging with high spatial resolution by acoustically detecting optical absorption contrasts via the photoacoustic effect. We developed a compact handheld OR-PAM probe for fast

Nicotinamide N‐methyltransferase promotes drug resistance in lung cancer, as revealed by nascent proteomic profiling

open access: yesMolecular Oncology, EarlyView.
AZD9291 has shown promise in targeted cancer therapy but is limited by resistance. In this study, we employed metabolic labeling and LC–MS/MS to profile time‐resolved nascent protein perturbations, allowing dynamic tracking of drug‐responsive proteins. We demonstrated that increased NNMT expression is associated with drug resistance, highlighting NNMT ...

Phase imaging with computational specificity (PICS) for measuring dry mass changes in sub-cellular compartments

open access: yesNature Communications, 2020
Quantitative phase imaging suffers from a lack of specificity in label-free imaging. Here, the authors introduce Phase Imaging with Computational Specificity (PICS), a method that combines phase imaging with machine learning techniques to provide ...
