Improving PARP inhibitor efficacy in bladder cancer without genetic BRCAness by combination with PLX51107

Clinical trials on PARP inhibitors in urothelial carcinoma (UC) showed limited efficacy and a lack of predictive biomarkers. We propose SLFN5, SLFN11, and OAS1 as UC‐specific response predictors. We suggest Talazoparib as the better PARP inhibitor for UC than Olaparib.

Methylation biomarkers can distinguish pleural mesothelioma from healthy pleura and other pleural pathologies

We developed and validated a DNA methylation–based biomarker panel to distinguish pleural mesothelioma from other pleural conditions. Using the IMPRESS technology, we translated this panel into a clinically applicable assay. The resulting two classifier models demonstrated excellent performance, achieving high AUC values and strong diagnostic accuracy.

Linear probabilistic sequential machines

Based on the notion of a stochastic matrix operator, the class of linear probabilistic sequential machines is defined and characterized. Necessary and sufficient conditions are formulated for a given probabilistic sequential machine to be linear. Next‐generation proteomics improves lung cancer risk prediction

This is one of very few studies that used prediagnostic blood samples from participants of two large population‐based cohorts. We identified, evaluated, and validated an innovative protein marker model that outperformed an established risk prediction model and criteria employed by low‐dose computed tomography in lung cancer screening trials.
