In Vitro Propagation of Rheophytic Orchid, Epipactis flava Seidenf.—A Comparison of Semi-Solid, Continuous Immersion and Temporary Immersion Systems

open access: yesBiology, 2019
Epipactis flava Seidenf. is an endangered Thai rheophytic orchid that has recently shown a rapid decrease in its natural habitat, prompting an urgent need for conservation using ex situ reintroduction methods.

Effect of Liquid Culture Systems (Temporary Immersion Bioreactor and Rotary Shaker) Used During Multiplication and Differentiation on Efficiency of Repetitive Somatic Embryogenesis of Narcissus L. ‘Carlton’

open access: yesAgronomy
Liquid culture systems, including bioreactors, are valuable tools for the scaling up of production. Their involvement leads to the automation of the highly efficient, reproducible somatic embryogenesis of Narcissus L. ‘Carlton’.

In vitro degradation of diacetoxyscirpenol and T-2 toxin by use of Mucor racemosus fresen. f. racemosus isolate [PDF]

open access: yesZbornik Matice Srpske za Prirodne Nauke, 2011
Under controlled in vitro conditions the capacity of the Mucor racemosus f. racemosus 1215/09 isolate to degrade type A trichothecenes (diacetoxyscirpenol - DAS and T-2 toxin) was observed in the liquid nutritive medium.

Effect of liquid media culture systems on yam plant growth (Dioscorea alata L. 'Pacala Duclos') [PDF]

open access: yesBiotechnologie, Agronomie, Société et Environnement, 2011
The culture system type in liquid media influences yam plant growth ('Pacala Duclos' clone). In culture systems with forced renewal of internal atmosphere in culture flasks, Temporary Immersion System (TIS) and Constant Immersion System (CIS) with ...

Spatiotemporal and quantitative analyses of phosphoinositides – fluorescent probe—and mass spectrometry‐based approaches

open access: yesFEBS Letters, EarlyView.
Fluorescent probes allow dynamic visualization of phosphoinositides in living cells (left), whereas mass spectrometry provides high‐sensitivity, isomer‐resolved quantitation (right). Their synergistic use captures complementary aspects of lipid signaling. This review illustrates how these approaches reveal the spatiotemporal regulation and quantitative

Calpain small subunit homodimerization is robust and calcium‐independent

open access: yesFEBS Letters, EarlyView.
Calpains dimerize via penta‐EF‐hand (PEF) domains. Using single‐molecule force spectroscopy, we measured the strength and kinetics of PEF–PEF homodimer binding. The interaction is robust, shows a transient conformational step before dissociation, and remains largely insensitive to Ca2+.

Structural insights into an engineered feruloyl esterase with improved MHET degrading properties

open access: yesFEBS Letters, EarlyView.
A feruloyl esterase was engineered to mimic key features of MHETase, enhancing the degradation of PET oligomers. Structural and computational analysis reveal how a point mutation stabilizes the active site and reshapes the binding cleft, expading substrate scope.

Valosin‐containing protein counteracts ATP‐driven dissolution of FUS condensates through its ATPase activity in vitro

open access: yesFEBS Letters, EarlyView.
Biomolecular condensates formed by fused in sarcoma (FUS) are dissolved by high ATP concentrations yet persist in cells. Using a reconstituted system, we demonstrate that valosin‐containing protein (VCP), an AAA+ ATPase, counteracts ATP‐driven dissolution of FUS condensates through its D2 ATPase activity.
