Targeting PLD3 Reverses the Immunosuppressive Niche by Reprogramming Tumor‐Associated Macrophages and Potentiates Antitumor Immunity

PLD3 activates the lysosomal‐AKT‐NF‐κB axis to drive cellular senescence in macrophages, establishing an immunosuppressive TME by limiting the infiltration of cytotoxic T, NK, and NKT cells, which confers resistance to anti‐PD‐1 therapy. Abrine inhibits PLD3 expression, restoring antitumor immunity and synergizing with anti‐PD‐1 treatment.

Noninvasive Characterization of Tumor Heterogeneity in HNSCC: From Clinical Utility to Biological Correlates

SSR4 sustains Tertiary Lymphoid Structures by Regulation Quality Control of N‐linked Glycosylation During B‐cell Differentiation Into Plasmacyte in Colorectal Cancer

SSR4, a TRAP component induced in B cells, governs BAFFR N‐glycosylation via DDOST to sustain NF‐κB signaling, B‐cell differentiation, and TLS maturation. A Non‐Canonical Role of SMAD4 in Regulating 3D Genome Architecture to Inhibit Lung Squamous Cell Carcinoma Development

SMAD4 is identified as a guardian of 3D genome architecture in lung squamous cell carcinoma. Loss of SMAD4 unleashes EP300 at chromatin loop anchors, strengthening enhancer–promoter looping and H3K27ac at the SOX2 locus to drive aberrant SOX2 activation and tumor cell proliferation.
