KRAS mRNA Spleen‐Targeting Lipid Nanoparticles Synergize with Irinotecan Silicasomes to Robustly Augment the Cancer Immunity Cycle in Pancreatic Cancer

open access: yesAdvanced Science, EarlyView.
To overcome pancreatic cancer's immunosuppressive barriers, this study explores a dual approach: triggering immunogenic tumor cell death at the primary tumor site with irinotecan‐loaded silicasomes and enhancing immune activation in the spleen using LNPs encapsulating tumor antigen mRNA and a TLR7/8 agonist.

The Immune Microenvironment: New Therapeutic Implications in Organ Fibrosis

open access: yesAdvanced Science, EarlyView.
This review summarizes recent advances in understanding the immune microenvironment's role in fibrosis, focusing on phenotypic/functional alterations of immune cells and their dynamic interactions with other cellular constituents within tissues. The authors further explore therapeutic opportunities and challenges in targeting immune microenvironment ...

Circulating Tumor Cells Shed Shearosome Extracellular Vesicles in Capillary Bifurcations That Activate Endothelial and Immune Cells

open access: yesAdvanced Science, EarlyView.
Using microfluidic models of capillary networks, this study discovered that fluid shear stresses on circulating tumor cells transiting through capillary‐sized bifurcations shed a novel class of shear‐dependent large extracellular vesicle, “shearosomes”, whose proteome comprises of over 3000 distinct proteins.

ROS‐Responsive Hydrogel for Localized Delivery of Nampt and Stat3 Inhibitors Exhibits Synergistic Antitumor Effects in Colorectal Cancer Through Ferroptosis Induction and Immune Microenvironment Remodeling

open access: yesAdvanced Science, EarlyView.
In this study, ferroptosis in colorectal cancer cells is induced by FK866 through the NAD⁺/STAT3/GPX4 pathway. To minimize systemic toxicity, a reactive oxygen species (ROS)‐responsive hydrogel is developed to co‐deliver FK866 and C188‐9, resulting in synergistic and localized antitumor effects.

A Novel Class of Multi‐substituted Diaryl Scaffold Derivatives Inhibit Glioblastoma Progression by Targeting CD155

open access: yesAdvanced Science, EarlyView.
Structural modification of clofoctol yielded a novel blood‐brain barrier‐permeable compound, B7, featuring a multi‐substituted diaryl scaffold. B7 demonstrates potent anti‐glioblastoma activity by suppressing glioblastoma stem cells proliferation, migration, and invasion through interactions with five key residues of poliovirus receptor cell adhesion ...
