Discovery of a Novel and Potent Kir4.1 Inhibitor as a Safe and Rapid‐Onset Antidepressant Agent in Mice

open access: yesAdvanced Science, EarlyView.
The preferred derivative JX3212 demonstrates strong inhibitory activity against Kir4.1 with favorable druggability and shows significant antidepressant efficacy in vivo. Abstract Major depressive disorder is a serious psychiatric disorder for which novel and fast‐acting antidepressants are required.

Integration of non-randomized studies with randomized controlled trials in meta-analyses of clinical studies: a meta-epidemiological study on effect estimation of interventions

open access: yesBMC Medicine
Backgrounds Syntheses of non-randomized studies of interventions (NRSIs) and randomized controlled trials (RCTs) are increasingly used in decision-making.

Construction of PANoptosis‐Inhibiting Carbonized Polymer Dots via Machine Learning Potential for Mitigating Chemodrug‐Induced Nephrotoxicity

open access: yesAdvanced Science, EarlyView.
An enhanced sampling method is developed for molecular dynamics based on trained machine learning force fields to guide the construction of PANoptosis‐inhibiting carbonized polymer dots (Lu‐CDs) derived from a flavonoid compound for treating chemodrug‐induced nephrotoxicity.

Impacts of COVID-19 pandemic on early life gut microbiome

open access: yesGut Microbes
Increased hygiene and sanitation are theorized to predispose to developing atopic diseases, a process potentially mediated by the gut microbiome. We hypothesized that the gut microbiome maturation has been altered by COVID-19 lockdown measures during the

Puerarin Targets MIC19 to Suppress Mitochondrial Metabolism of Tumor‐Infiltrating Tregs and Enhance Anti‐tumor Immunity

open access: yesAdvanced Science, EarlyView.
Puerarin targets mitochondrial MIC19 in tumor‐infiltrating Tregs, impairing their immunosuppressive function and enhancing antitumor immunity. This study uncovers the metabolic dependency of tumor‐infiltrating Tregs and highlights a potential metabolic‐targeted immunotherapeutic strategy for liver cancer. Abstract Regulatory T cells (Tregs) are pivotal

A Dynamic Gate Enables Regioselective Hydroxylation of Free Arginine by a Non‐Canonical Heme Enzyme

open access: yesAdvanced Science, EarlyView.
The YqcI/YcgG enzyme AglA employs a unique “sandwich” fold and a dynamic gating mechanism to selectively hydroxylate AMPn‐Arg at Nε. Site‐directed mutations reprogram regioselectivity or enable activity on free arginine by altering hydrogen‐bond networks, precisely positioning substrates.

scPER: A Rigorous Computational Approach to Determine Cellular Subtypes in Tumors Aligned With Cancer Phenotypes From Total RNA Sequencing

open access: yesAdvanced Science, EarlyView.
scPER presents an adversarial‐autoencoder framework that deconvolves bulk total RNA‐seq to quantify tumor‐microenvironment cell types and uncover phenotype‐linked subclusters. Across diverse benchmarks, scPER improves accuracy over existing tools.

Paradoxical Topological Soliton Lattice in Anisotropic Frustrated Chiral Magnets

open access: yesAdvanced Science, EarlyView.
The article describes the discovery of a stable skyrmion‐antiskyrmion lattice (S‐AL) in anisotropic frustrated chiral magnets. This lattice has a net‐zero topological charge due to a balanced population of skyrmions and antiskyrmions. This is a paradoxical finding since these particles normally annihilate each other.
