Mannanase production by the lettuce endosperm

Endo-β-mannanase (EC 3.2.1.78) is produced and secreted by the cells of the endosperm of lettuce (lactuca sativa L.) "seeds" (achenes). In imbibed intact seeds, production is prevented by inhibitors. If the endosperm is incubated alone, these inhibitors can be removed by leaching, allowing mannanase production.

β-MANNANASES OF FUNGI

β-1,4-Mannanases are of common occurrence in fungi. They are induced enzymes, the yields of enzyme being 10- to 100-fold greater when the organism is grown on mannan. All of the β-mannanases studied are endopolysaccharases acting on long chains in a random manner and unable to act on mannotriose or mannobiose.

Purification and properties of mannanase from Bacillus subtilis

Extracellular mannanase from Bacillus subtilis NM-39, an isolate from Philippine soil, was purified about 240-fold with a yield of 7.3% by ammonium sulphate fractionation, DEAE-Toyopearl chromatography and Sephacryl S-200 gel filtration. Its M r was 38 kDa and it had a pI of 4.8 and optimum activity at pH 5.0 and 55°C.
