Variant Class–Specific Phenotypes and Outcomes in Pediatric TBX4‐Associated Pulmonary Hypertension
ABSTRACT Aim To describe the variant class associations with phenotype, natural history, treatment response, and outcomes in children with TBX4‐associated pulmonary hypertension (TBX4‐PH). Methods Retrospective cohort analysis of children < 18 years referred to the National Paediatric Pulmonary Hypertension Service between 2001 and 2025. Results Twenty‐

ABSTRACT Background and Purpose Prolonged weaning—characterized by failure in three consecutive spontaneous breathing trials (SBTs) and requiring over 7 days of mechanical ventilation post‐initial SBT—presents a critical clinical challenge often exacerbated by intensive care unit‐acquired weakness (ICU‐AW) and impaired airway clearance.

ABSTRACT Accurate prediction of time‐to‐event outcomes is essential for clinical decision‐making and model‐informed drug development (MIDD). Conventional survival models may inadequately represent complex longitudinal patient trajectories, time‐dependent treatments, and evolving biomarker or pharmacokinetic/pharmacodynamic (PK/PD) profiles.

Early predictors of persistent childhood asthma and allergic rhinitis: A machine learning study using post‐hoc explainability [PDF]
Machine learning models applied to detailed early‐life data showed limited ability to predict which children developed persistent asthma or allergic rhinitis. Although clinically plausible features contributed to predictions, model performance was insufficient for reliable individual prognostic stratification.

Summary Introduction Facemask ventilation is a key airway management skill but predicting difficulty can be challenging. Pre‐operative three‐dimensional face scanning may have diagnostic value. We aimed to identify interpretable facial shape features and to quantify their value for predicting difficult facemask ventilation.
