Nuclear factor of activated T cells as a driver of tumor progression and a target for precision therapy

open access: yesAnimal Models and Experimental Medicine, EarlyView.
Overview illustration of dysregulated NFAT activation coordinating upstream signaling, post‐translational regulation and cancer promoting pathways to induce abnormal gene expression that drives tumor development, progression and immune escape. Abstract The Nuclear Factor of Activated T Cells (NFAT) family comprises closely related transcription factors.

Taking Diabetes to Heart—Deregulation of Myocardial Lipid Metabolism in Diabetic Cardiomyopathy [PDF]

open access: yesJournal of the American Heart Association, 2013
Heart disease is the leading cause of death in patients with diabetes.[1][1] Although advances in medical management and lifestyle interventions have reduced cardiovascular mortality in diabetic patients by as much as 40% over the last decade, the actual number of deaths is predicted to rise as ...

The Nuclear Receptor Testicular Receptor 4 Orchestrates Cytoskeletal Organization in a G Protein Subunit Alpha 12/Rho‐Associated Protein Kinase–Dependent Manner to Promote Myofibroblast Differentiation and Tissue Fibrosis in Systemic Sclerosis

open access: yesArthritis &Rheumatology, EarlyView.
Objective Members of the superfamily of nuclear receptors have been implicated in inflammatory processes and pathologic tissue remodeling and have emerged as attractive targets for pharmaceutical intervention. However, the role of testicular receptor 4 (TR4; or Nr2c2) in fibroblast activation and rheumatologic diseases has not yet been investigated ...

Efficacy, safety and cost‐effectiveness of CAR‐T therapy

open access: yesBritish Journal of Clinical Pharmacology, EarlyView.
CAR T‐cells demonstrate high efficacy in blood cancers, including ALL, MM and DLBCL. Innovations target solid tumours despite challenges such as antigen escape. Combination therapies enhance the delivery and infiltration of CAR T cells. Toxicity, cost and resistance remain major barriers to clinical use.

From anthelmintic to neuro‐oncology: A systematic review of mebendazole repurposing for brain tumour therapy

open access: yesBritish Journal of Clinical Pharmacology, EarlyView.
Abstract Aim Mebendazole (MBZ), a benzimidazole anthelmintic with established clinical use, has emerged as a repurposing candidate for primary brain tumours due to its multimodal anticancer actions and central nervous system penetrance. This systematic review synthesizes preclinical and clinical evidence evaluating MBZ's efficacy, mechanisms of action ...

Compound heterozygous SLC12A5 variants expand the molecular and functional spectrum of KCC2‐developmental and epileptic encephalopathy

open access: yesEpilepsia, EarlyView.
Overview of the multimodal experimental approach integrating clinical, genetic, in silico, and in vitro investigations. Clinical: Representative EEG recording setup and ictal traces from affected patients. Genetic: Pedigrees for Families A and B highlighting the inheritance of the four identified SLC12A5 variants (A1, A2, B1, B2).

Genetics and metabolic deregulation following cancer initiation: A world to explore

open access: yesBiomedicine & Pharmacotherapy, 2016
Cancer is a group of highly complex and heterogeneous diseases with several causes. According to the stochastic model, cancer initiates from mutation in somatic cells, leading to genomic instability and cell transformation. This canonical pathway of carcinogenesis is related to the discovery of important mechanisms that regulate cancer initiation ...

Syncing Health Timing: Exploring the Interplay of Circadian Rhythms, Obesity, and Cardiovascular Diseases

open access: yesFood Safety and Health, EarlyView.
Circadian rhythm disruption impairs metabolic regulation, appetite control, and vascular function, promoting obesity and cardiovascular diseases. Evidence highlights a bidirectional relationship where metabolic disorders further disturb circadian timing.

Adipose Tissue Integrity as a Prerequisite for Systemic Energy Balance

open access: yes, 2007
Peroxisome proliferator-activated receptor gamma (PPARgamma) is an essential regulator of adipocyte differentiation, maintenance, and survival. Deregulations of its functions are associated with metabolic diseases.
