Procyanidin B3 and Its Derivatives Alleviate Neuronal Injury by Targeting G3BP1 for Ischemic Stroke Therapy

open access: yesAdvanced Science, EarlyView.
Ischemic stroke is a serious disease with high rates of mortality and disability, but there is a lack of novel therapeutic targets and agents for it. Now it is shown that Ras GTPase‐activating protein SH3 domain‐binding protein 1 (G3BP1) has the potential to serve as a therapeutic target for ischemic stroke.

Energy Deficiency‐Induced ATG4B Nuclear Translocation Inhibits PRMT1‐Mediated DNA Repair and Promotes Leukemia Progression

open access: yesAdvanced Science, EarlyView.
This study demonstrated that ATG4B translocates from the cytoplasm to the nucleus during energy deficiency. Nuclear ATG4B inhibits DNA repair by interacting with PRMT1. The nuclear ATG4B‐mediated DNA repair defect is significantly exacerbated within acute myeloid leukemia (AML) cells, promoting leukemia progression in an AML mouse model.

Hypoxia‐Induced PRMT1 Lactylation Drives Vimentin Arginine Asymmetric Dimethylation in Tumor Metastasis

open access: yesAdvanced Science, EarlyView.
Hypoxia triggers lactylation of PRMT1 at K134/K145, enhancing its methyltransferase activity to asymmetrically dimethylate vimentin R64. This post‐translational cascade promotes vimentin filament assembly, cytoskeletal remodeling, and tumor metastasis, with PRMT1 inhibition (MS023) reducing metastatic colonization.

UHRF1 Controls the Timing of RAD51 Removal During DNA Damage Repair Through Suppressing RFWD3

open access: yesAdvanced Science, EarlyView.
We demonstrate here that the recombinase RAD51 is protected by UHRF1 through ubiquitinating RFWD3, an E3 for RAD51, and this very process is inhibited by RAD51, leading to a triple negative feedback circuit that ensures appropriate levels of RFWD3 and RAD51 for DNA damage response. Abstract The RAD51 recombinase is evolutionarily conserved critical for

Targeted Degradation of sGRP78 Alleviates the Immunosuppressive Tumor Microenvironment

open access: yesAdvanced Science, EarlyView.
Soluble GRP78 (sGRP78) is identified as a novel tumor‐derived soluble factor that induces regulatory T cells (Treg). sGRP78 serves as an excellent serological biomarker for predicting the efficacy of neoadjuvant therapy in breast cancer patients. sGRP78 specifically binds to B cells, inducing the expression of IL‐10 and PD‐L1, thereby promoting Treg ...
