Spatiotemporal and quantitative analyses of phosphoinositides – fluorescent probe—and mass spectrometry‐based approaches

open access: yesFEBS Letters, EarlyView.
Fluorescent probes allow dynamic visualization of phosphoinositides in living cells (left), whereas mass spectrometry provides high‐sensitivity, isomer‐resolved quantitation (right). Their synergistic use captures complementary aspects of lipid signaling. This review illustrates how these approaches reveal the spatiotemporal regulation and quantitative

An intracellular transporter mitigates the CO2‐induced decline in iron content in Arabidopsis shoots

open access: yesFEBS Letters, EarlyView.
This study identifies a gene encoding a transmembrane protein, MIC, which contributes to the reduction of shoot Fe content observed in plants under elevated CO2. MIC is a putative Fe transporter localized to the Golgi and endosomal compartments. Its post‐translational regulation in roots may represent a potential target for improving plant nutrition ...

A compact two-wave dichrometer of an optical biosensor analytical system for medicine

open access: yesEPJ Web of Conferences, 2017
An experimental model has been developed of a compact two-wave dichrometer on the base of LEDs that is well-suited to work with “liquid” DNA nanoconstructions as biosensing units.

The Caenorhabditis elegans DPF‐3 and human DPP4 have tripeptidyl peptidase activity

open access: yesFEBS Letters, EarlyView.
The dipeptidyl peptidase IV (DPPIV) family comprises serine proteases classically defined by their ability to remove dipeptides from the N‐termini of substrates, a feature that gave the family its name. Here, we report the discovery of a previously unrecognized tripeptidyl peptidase activity in DPPIV family members from two different species.

Nanoconstructions on the base of double-stranded DNA molecules and their optical properties

open access: yesEPJ Web of Conferences, 2017
Experimental results have been presented on studying optical properties of nanoconstructions formed of orientationally ordered neighbouring double-stranded DNA molecules in the structure of their liquid-crystalline phases and dispersion particles of ...

Peptide‐based ligand antagonists block a Vibrio cholerae adhesin

open access: yesFEBS Letters, EarlyView.
The structure of a peptide‐binding domain of the Vibrio cholerae adhesin FrhA was solved by X‐ray crystallography, revealing how the inhibitory peptide AGYTD binds tightly at its Ca2+‐coordinated pocket. Structure‐guided design incorporating D‐amino acids enhanced binding affinity, providing a foundation for developing anti‐adhesion therapeutics ...

Array detection enables large localization range for simple and robust MINFLUX

open access: yesLight: Science & Applications
The MINFLUX concept significantly improves the localization properties of single-molecule localization microscopy (SMLM) by overcoming the limit imposed by the fluorophore’s photon counts.

Laser scanning reflection-matrix microscopy for aberration-free imaging through intact mouse skull

open access: yesNature Communications, 2020
Microscopic imaging of the brain usually requires thinning of the skull, as it causes complex aberration. Here, the authors introduce a label-free imaging modality termed laser scanning reflection-matrix microscopy, which allows for correcting these ...

Real‐time assay of ribonucleotide reductase activity with a fluorescent RNA aptamer

open access: yesFEBS Letters, EarlyView.
Ribonucleotide reductases (RNR) synthesize DNA building blocks de novo, making them crucial in DNA replication and drug targeting. FLARE introduces the first single‐tube real‐time coupled RNR assay, which enables isothermal tracking of RNR activity at nanomolar enzyme levels and allows the reconstruction of allosteric regulatory patterns and rapid ...

In situ and real-time ultrafast spectroscopy of photoinduced reactions in perovskite nanomaterials

open access: yesNature Communications
By employing two synchronized mode-locked femtosecond lasers and interferometric detection of the pump-probe spectra—referred to as asynchronous and interferometric transient absorption—we have developed a method for broad dynamic range and rapid data ...
