mTOR Dysregulation, Insulin Resistance, and Hypertension

open access: yesBiomedicines
Worldwide, diabetes mellitus (DM) and cardiovascular diseases (CVDs) represent serious health problems associated with unhealthy diet and sedentarism. Metabolic syndrome (MetS) is characterized by obesity, dyslipidemia, hyperglycemia, insulin resistance (

Comprehensive In Vitro Profiling Demonstrates Successful Immune Evasion of Bioengineered Immune‐Shielded Heart Valves

open access: yesAdvanced Healthcare Materials, EarlyView.
Living donor heart valves offer superior durability for pediatric patients but trigger immune rejection. Lentiviral delivery of viral immune‐evasion genes US2 and Serpin b9 into donor valve cells and valve tissue suppresses HLA expression, reduces PBMC clustering and cytotoxic killing, and induces transcriptional immune‐tolerance programs, paving the ...

Inhibition of PI3K/AKT/mTOR signaling enhances autophagy in HL-60 acute myeloid leukemia cells: An integrative bioinformatic and in vitro study

open access: yesBiochemistry and Biophysics Reports
Background: Acute myeloid leukemia (AML) involves uncontrolled proliferation of myeloid progenitor cells and carries a poor prognosis. The PI3K/AKT/mTOR pathway plays a key role in AML pathogenesis by regulating cancer cell proliferation and survival ...

Advanced Materials for Biologics Delivery to Brain Tumors

open access: yesAdvanced Materials, EarlyView.
Material innovation is central to unlocking the therapeutic potential of biologics against many central nervous system diseases, including brain cancer. By engineering carriers with controlled transport, targeting, and release properties, advanced materials can overcome the blood–brain barrier and tumor microenvironment, improving the delivery of ...

Abstract IA25: Third-generation inhibitors of mTOR

open access: yes, 2017
Precision medicines exert selective pressure on tumor cells that leads to the preferential growth of resistant subpopulations, necessitating the development of next-generation therapies to treat the evolving cancer.

Rab1A Promotes Hepatic Steatosis by Suppressing Mitophagy via the Raf‐1/ERK1/2/PINK1 Signaling Axis

open access: yesAdvanced Science, EarlyView.
Why does fat accumulate in the liver? Our study reveals Rab1A as the molecular switch that silences the cell's mitochondrial cleanup crew. Turning Rab1A off reactivates mitophagy, clears excess fat, and reverses fatty liver disease in mice—offering a promising new therapeutic target for MASLD, a growing global health challenge.

Innate Immunocompetent hiPSC‐Derived Neurospheroids Capture Early CNS Responses to rAAV

open access: yesAdvanced Science, EarlyView.
Knowledge of human CNS immune responses to AAV‐based gene therapies remains limited due to the lack of immune‐competent human models. Here, a hiPSC‐derived 3D neuroimmune platform integrating neurospheroids and microglia is established using stirred‐tank bioreactors.

A Critical Time Window of Neocortex Modulation for Remote Fear Memory in Rats and Humans

open access: yesAdvanced Science, EarlyView.
This study identifies a delayed consolidation window around 18 h after fear learning in the mPFC essential for remote fear memory. VTA dopaminergic input during this window sustains remote memory in rats, while non‐invasive stimulation of homologous human mPFC at the corresponding time selectively weakens remote but not recent fear memory, revealing a ...

Engineering Approaches to Modify Immunomodulatory Functions of Mesenchymal Stromal Cells (MSCs): Tissue Regeneration and Clinical Application

open access: yesAdvanced Science, EarlyView.
Mesenchymal stromal cells (MSCs) show promise for treating immune‐related disorders through immunomodulation and tissue regeneration. This review gives a brief overview of current clinical approval of MSC therapies. It also discussed how bioengineering, including genetic modification, biomaterial delivery, extracellular vesicles, and iPSC‐derived MSCs,
