Advanced Science, EarlyView.FAK orchestrates a mechanical barrier limiting nanomedicine accumulation in solid tumors by coupling ECM remodeling with cytoskeletal tension. Genetic or pharmacological FAK inhibition dismantles this barrier, enhances clathrin‐mediated uptake and penetration, and overcomes delivery limitations to improve antitumor efficacy.
